摘 要:目的:建立并评价符合中医证候特点的脾虚湿盛肝郁证溃疡性结肠炎(ulcerative colitis,UC)大鼠模型。方法:45只SPF级SD大鼠随机分为正常对照组、葡聚糖硫酸钠(DSS)组、病证结合组、模型组。正常对照组正常饲养,DSS组定量灌胃10%DSS溶液,病证结合组采用环境、饮食、情绪干预造模,模型组采用DSS组结合病证结合组方法造模。观察各组大鼠一般情况及病理改变,酶联免疫吸附测定法(ELISA)检测大鼠血清中白细胞介素-2(IL-2)和肿瘤坏死因子-α(TNF-α)含量,光镜下观察大鼠结肠组织HE染色切片。结果:与正常对照组比较,模型组大鼠体质量显著降低,UC活动指数评分(DAI)及结肠黏膜损伤指数(CMDI)显著升高,血清IL-2含量显著降低,TNF-α含量显著升高(P<0.01),光镜下观察黏膜层溃烂严重,腺体破坏,大量炎症细胞浸润,间质水肿、充血明显。结论:采用DSS反复诱导结合饮食、环境干预制备的脾虚湿盛肝郁证UC大鼠模型,符合中医证候特点和UC病理特点,是较理想的脾虚湿盛肝郁证UC模型。Objective: To establish and evaluate a rat model of ulcerative colitis(UC) with the syndrome of spleen deficiency, dampness and liver stagnation. Methods: A total of 45 SPF SD rats were randomly divided into normal control group, dextran sodium sulfate(DSS) group, disease and syndrome combination group, and model group. The normal control group was fed normally, the DSS group was given 10% DSS solution quantitatively, the disease and syndrome combination group was modeled by environment, diet, and emotion intervention,and the model group was modeled by the DSS group and disease and syndrome combination group. The general conditions and pathological changes of the rats in each group were observed. The contents of interleukin 2(IL-2)and tumor necrosis factor(TNF-α) in serum of rats were detected by enzyme-linked immunosorbent assay(ELISA). HE staining sections of the colon tissues of the rats were observed under the microscope. Results:Compared with the normal control group, the body mass of rats in the model group was significantly reduced,the UC Activity Index(DAI) and Colonic Mucosal Injury Index(CMDI) were significantly increased, the serum IL-2 content was significantly decreased, and the TNF-α content was significantly increased(P <0.01). Under light microscope, the mucosal layer was severely ulcerated, glands were destroyed, a large number of inflammatory cells infiltrated, interstitial edema and congestion were obvious. Conclusion: The UC rat model of spleen defi-ciency, dampness and liver-stagnation syndrome prepared by DSS repeated induction combined with diet and environmental intervention is in line with the characteristics of TCM syndromes and pathological characteristics of UC. It is an ideal UC model of spleen deficiency, dampness and liver stagnation syndrome.
