摘 要:目的:建立重芪结肠靶向微丸(PA-CTPC)中黄芪总多糖和重楼皂苷Ⅰ、Ⅱ、Ⅶ、H的含量测定方法;评价PA-CTPC对葡聚糖硫酸钠(DSS)诱导的溃疡性结肠炎(UC)小鼠的干预作用。方法:采用苯酚-浓硫酸法测定PA-CTPC中黄芪总多糖含量;采用高效液相色谱法(HPLC)测定重楼皂苷Ⅰ、Ⅱ、Ⅶ、H的含量;KM雄性小鼠随机分为正常组、模型组和PA-CTPC高、中、低剂量组(0.4、0.2、0.1 mg/g),采用4.0%DSS溶液自由饮用诱导建立小鼠UC模型,通过疾病活动指数(DAI)评分、结肠组织形态、病理损伤评分及血清白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)、白细胞介素-10(IL-10)、白细胞介素-4(IL-4)、D-乳酸(D-LA)、二胺氧化酶(DAO)水平评价PA-CTPC对DSS诱导的小鼠UC模型的干预作用。结果:苯酚-浓硫酸法测定3批PA-CTPC中黄芪总多糖平均含量分别为158.33、151.20、156.70 mg/g;HPLC法测定3批PA-CTPC中重楼皂苷Ⅰ、Ⅱ、Ⅶ、H的平均总含量分别为49.56、50.00、50.13 mg/g;PA-CTPC中、高剂量组可明显改善UC模型小鼠一般状况,降低DAI评分、结肠大体形态和组织病理学评分,明显降低血液中IL-6、TNF-α、D-LA、DAO水平,升高IL-10、IL-4水平。结论:建立的PA-CTPC中黄芪总多糖和重楼皂苷Ⅰ、Ⅱ、Ⅶ、H的含量测定方法准确、稳定、可行,可用于PA-CTPC的质量控制。PA-CTPC对DSS诱导的小鼠溃疡性结肠炎具有治疗作用,与调控炎性因子表达,改善肠道黏膜屏障功能有关。Objective: To established the content determination method of Astragalus Total Polysaccharide and Paris Polyphylla Saponin Ⅰ, Ⅱ, Ⅶ and H in PA-CTPC, and to evaluate the intervention effect of PA-CTPC on DSS induced UC in mice. Methods: Astragalus Total Polysaccharide in the PA-CTPC was determined by phenol-concentrated sulfuric acid method and Paris Polyphylla Saponin Ⅰ, Ⅱ, Ⅶ and H was determined by HPLC. KM male mice were randomly divided into normal group, model group and PA-CTPC high, medium and low groups(0.4, 0.2 and 0.1 mg/g), then mice model of UC was established by free drinking of 4.0% DSS solution.The intervention effect of PA-CTPC on DSS induced mice model of UC was evaluated by DAI, colon morphology and histopathology scores and levels of IL-6, TNF-α, IL-10, IL-4, D-LA and DAO. Results: The average contents of Astragalus Total Polysaccharides determined by phenol-concentrated sulfuric acid method were 158.33, 151.20 and 156.70 mg/g respectively in the three batches of PA-CTPC. And the Paris Polyphylla Saponin Ⅰ, Ⅱ, Ⅶ and H determined by HPLC was respectively 49.56, 50.00 and 50.13 mg/g;PA-CTPC medium and high groups can significantly improve the general condition of UC model mice, reduce scores of DA, colonic morphology and histopathological, decrease levels of IL-6, TNF-α, D-LA and DAO in blood, and increase levels of IL-10 and IL-4. Conclusion: The established method for the determination of Astragalus Total Polysaccharide and Paris Polyphylla Saponin Ⅰ, Ⅱ, Ⅶ and H in PA-CTPC is accurate, stable and feasible, which can be used for the quality control of PA-CTPC. PA-CTPC shows a therapeutic effect on DSS induced UC in mice, which is related to regulating the expression of inflammatory factors and improving the intestinal mucosal barrier function.
