摘  要:目的探讨miRNA-138(miR-138)及其相关靶蛋白细胞周期蛋白D1(Cyclin D1)、P53在皮肤扁平苔藓(LP)中的表达。方法收集活检切取的LP病人皮损组织标本40例(LP组),选取同期美容外科手术切除的正常皮肤组织标本35例作为对照组,采用RT-PCR法检测两组标本miR-138表达,ELISA法检测Cyclin D1、P53蛋白表达。应用Spearman秩相关检验分析miR-138表达与Cyclin D1、P53表达之间的相关性。结果与对照组比较,LP组Cyclin D1、P53蛋白表达明显升高(t=-2.257、-2.217,P<0.05),miR-138表达下调(t=2.473,P<0.05)。Spearman秩相关分析显示,LP皮损组织miR-138表达与Cyclin D1、P53呈负相关(r=-0.248、-0.264,P<0.05)。结论miR-138可能在皮肤LP的发病机制中起重要作用。Objective To investigate the expression of miRNA-138(miR-138)and its related target proteins cyclin D1 and P53 in skin lichen planus(LP).Methods Forty skin lesion tissue specimens were collected from biopsies in LP patients(LP group),and 35 normal skin tissue specimens from cosmetic surgeries in the same period were collected as control group.RT-PCR was used to determine the expression of miR-138 in the two groups,and ELISA was used to determine the expression of cyclin D1 and P53 proteins.The Spearman rank correlation test was used to analyze the correlation of miR-138 expression with cyclin D1 and P53 expression.Results Compared with the control group,the LP group had significantly increased expression of cyclin D1 and P53 proteins(t=-2.257 and-2.217,P<0.05)and significantly down-regulated expression of miR-138(t=2.473,P<0.05).The Spearman rank correlation analysis showed that miR-138 expression in the skin lesion tissue of LP was negatively correlated with cyclin D1 and P53(r=-0.248 and-0.264,P<0.05).Conclusion miR-138 may play a key role in the pathogenesis of skin LP.
