摘 要:目的:补肾化痰方对肾虚模型裸鼠乳腺癌骨转移的影响。方法:将30只雌性裸鼠随机分为对照组、模型组及治疗组[补肾化痰方(3.67 g·kg-1)],每组10只。模型组及治疗组皮下注射氢化可的松制成肾虚模型。肾虚模型制成3 d后,以左心室注射肿瘤细胞法制成骨转移模型。对比3组裸鼠体质量变化、瘤体质量、骨转移部位数、骨转移组织甲状旁腺激素相关蛋白(recombinant parathyroid hormone related protein,PTHrP)、转化生长因子β(recombinant transforming growth factor beta,TGF-β)、骨唾液酸蛋白(bonesialoprotein,BSP)的表达。结果:与造模前比较,对照组造模后体质量显著增加(P<0.01),模型组造模后体质量明显降低(P<0.05),治疗组体质量无明显变化(P>0.05);与模型组比较,治疗组瘤质量明显减轻,骨转移部位数明显减少,骨组织中PTHrP、TGF-β、BSP表达明显降低(P<0.05)。结论:补肾化痰方减轻乳腺癌骨转移的机制可能与调节PTHrP、TGF-β、BSP的表达有关。Objective:To observe the influence of Decoction for Tonifying Kidney and Resolving Phlegm on bone metastasis of breast cancer in nude mice of kidney deficiency model.Methods:A total of 30 Jian′ou female nude mice were randomly divided into the control group,the model group and the treatment group [Decoction for Tonifying Kidney and Resolving Phlegm(3.67 g · kg-1)],with 10 mice in each group.The model group and the treatment group were subcutaneously injected with hydrocortisone to make kidney deficiency model.Three days after the kidney deficiency model was established,the model of bone metastasis was established by injecting tumor cells into the left ventricle.The changes of body weight,tumor weight,the number of metastases,and the expressions of parathyroid hormone related protein(PTHrP),transforming growth factor beta(TGF-β) and bone sialoprotein(BSP) in bone metastases were compared among the three groups.Results:Compared with before modeling,the body weight of the control group increased significantly(P<0.01),the body weight of the model group decreased significantly(P<0.05),and the body weight of the treatment group did not change significantly(P>0.05).Compared with the model group,the tumor weight,the number of bone metastases and the expressions of PTHrP,TGF-β and BSP in bone tissue significantly reduced in the treatment group(P<0.05).Conclusion:Kidney deficiency can aggravate bone metastasis of breast cancer.The mechanism of Decoction for Tonifying Kidney and Resolving Phlegm in reducing bone metastasis of breast cancer may be related to regulating the expressions of PTHrP,TGF-β and BSP.
