摘  要:目的探讨奥司他韦对寒冷刺激致鼻黏膜免疫屏障功能低下(MNMBD)模型小鼠的保护作用。方法将38只SPF级昆明种小鼠随机分为正常组(12只,等体积蒸馏水)、模型组(12只,等体积蒸馏水)和奥司他韦组(14只,奥司他韦40 mg/kg)。灌胃给药,每日1次,连续3次。末次给药30 min后,模型组和奥司他韦组小鼠置-20℃2 h,再转移至25℃30 min,循环刺激2次以建立寒冷刺激致鼻黏膜免疫屏障功能低下模型小鼠;正常组小鼠不予任何刺激。采用免疫印迹(Western Blot)法检测小鼠鼻黏膜组织免疫球蛋白A(IgA)、干扰素λ(IFN-λ)、干扰素λ受体1(IFNLR1)、白细胞介素10Rβ(IL-10Rβ)、干扰素调节因子9(IRF9)和黏病毒抗性蛋白1(Mx-1)表达水平。结果与正常组比较,模型组小鼠IgA,IFN-λ,IFNLR1,IL-10Rβ,IRF9,Mx-1蛋白表达水平均显著下降(P<0.05);与模型组比较,奥司他韦组小鼠IgA,IFN-λ,IL-10Rβ,IRF9,Mx-1蛋白表达水平均显著上升(P<0.05)。结论奥司他韦可提高小鼠鼻黏膜免疫屏障功能,其作用与促进小鼠鼻黏膜组织中IgA蛋白表达和激活IFN-λ信号通路相关。Objective To investigate the protective effect of Oseltamivir on the mouse nasal mucosal immune barrier dysfunction(MNMBD)induced by cold stimulation.Methods Totally 38 SPF Kunming mice were randomly divided into normal group(12 mice,equal volume of distilled water),model group(12 mice,equal volume of distilled water)and Oseltamivir group(14 mice,40 mg/kg of Oseltamivir).The mice were given intragastric administration once a day for three times.30 min after the last administration,the mice in the model group and Oseltamivir group were placed at-20℃for 2 h,then they were transferred to 25℃for 30 min,and were circularly stimulated twice to establish the MNMBD model induced by cold stimulation,the mice in the normal group were not stimulated.The expression levels of immunoglobulin A(IgA),interferon-λ(IFN-λ),interferon-λreceptor 1(IFNLR1),interleukin-10 Rβ(IL-10 Rβ),interferon regulatory factor 9(IRF9)and myxovirus resistance protein-1(Mx-1)in the nasal mucosa tissues of mice were detected bywestern blot.Results The protein expression of Ig A,IFN-λ,IFNLR1,IL-10 Rβ,IRF9 and Mx-1 in the model group were significantly lower than that in the normal group(P<0.05).The protein expressions of Ig A,IFN-λ,IL-10 Rβ,IRF9 and Mx-1 in Oseltamivir group were significantly higher than that in the model group(P<0.05).Conclusion Oseltamivir can improve the immune barrier function of nasal mucosal tissues of mice,which is related to promoting the protein expression of Ig A in nasal mucosa tissues and activating IFN-λsignal pathway.
