摘 要:目的:通过观察大鼠脊髓内治疗前后γ-氨基丁酸(Gamma-aminobutyric Acid,GABA)含量的变化,试图从分子水平探讨温针灸改善脊髓损伤后肌痉挛状态的机理。方法:将75只大鼠随机平分为5组,即A、B、C、D、E组。采用秦氏模型法造模,A组不做处理;B组行单纯椎板切除术;C、D、E组造模。脊髓损伤后肌痉挛模型成功后开始分别对D组与E组进行电针及温针灸治疗,A、B、C组只作捆绑固定,每天1次,每次30 min,连续14 d。分别在造模后、治疗前及治疗后进行Basso Beattie Bresnahan (BBB)运动功能评分及Ashworth肌张力评定量表评级。分别于治疗前各组处死3只及治疗后处死所有大鼠,取整段脊髓样本送检,测定GABA的含量。结果:治疗后,D组和E组大鼠双后肢BBB运动功能评分增加,脊髓内GABA含量提高,但与C组比较均无显著差异;C、D、E三组大鼠双后肢升高的肌张力未见降低;D组与E组大鼠双后肢肌张力比较无显著差异。结论:实验结果与假设不符,原因可能为样本数较少,疗程短;或通过其他通路而发挥作用。Objective: By observing the changes of GABA content in spinal cord of rats, to explore the mechanism of improving the muscle spasm after spinal cord injury by warm acupuncture from the molecular level. Methods: 75 rats were randomly divided into 5 groups, namely group A, group B, group C, group D and group E. Group A was not treated, group B underwent simple laminectomy, and group C, D and E were modeled by Qin’s model method. After the spinal cord injury and the muscle spasm model successfully, group D and E were treated by electroacupuncture and warm acupuncture, separately. Group A, B, and C were only bundled and fixed. BBB exercise capacity score and Ashworth rating were performed after modeling, and before and after treatment. Three rats were sacrificed in each group before treatment and all rats were sacrificed after treatment. The entire spinal cord samples were taken for examination to determine the GABA content. Results: After treatment, the BBB exercise capacity score of both hind limbs were increased and the GABA content in the spinal cord was increased in group D and group E, and there was no difference in group C. There was no decrease in the muscle tone of two hind limbs in the C, D and E group. There was no difference in muscle tone between the hind legs in group D and E. Conclusion: The experimental results are not consistent with the hypothesis, the reasons may be the number of samples was small and the treatment course was short, or other channels played an important role during the treatment.
