核盘菌对观赏向日葵防卫相关基因表达的影响

摘  要:以观赏向日葵(Helianthus annuus L.)中一个矮向日葵品种为试材,通过对四叶期观赏向日葵进行核盘菌诱导,采用提取RNA进行反转录、PCR荧光定量等方法研究其防卫相关基因的表达,以期探究核盘菌时观赏向日葵防卫相关基因表达的影响。结果表明:核盘菌侵染对观赏向日葵水杨酸(SA)信号路径中的关键基因NPR1、PR1表达量发生上调;ET介导信号传递路径中关键基因PDF1.2表达量有所升高;JA/ET途径的关键"节"点基因MPK4、EDS1表达量上调变化;NPR1、PR1、EDS1、PDF1.2和MPK4均明显上调表达,由此表明,观赏向日葵受SA和JA信号的调控,核盘菌对观赏向日葵相关防卫基因表达有一定的影响。A dwarf sunflower variety from ornamental sunflower(Helianthus annuus L.)was used as the test material.By inducing Sclerotinia sclerotiorum to the four-leaf stage ornamental sunflower,RNA was extracted for reverse transcription,PCR fluorescence quantitative methods to study the expression of defense-related genes and to investigate the effect of pantococcus radiodurans on the expression of defense-related genes in ornamental sunflowers.The results showed that Sclerotinia sclerotioruminfection upregulated the expression levels of key genes NPR1 and PR1 in the SA signaling pathway of ornamental sunflowers;the expression level of key genes PDF1.2 in the ETmediated signal transmission pathway increases;JA the expression of the key"node"genes MPK4 and EDS1 in the ET pathway was up-regulated;NPR1,PR1,EDS1,PDF1.2 and MPK4 were significantly up-regulated,indicating that ornamental sunflower is regulated by SA and JA signals,and S.sclerotiorum had a certain influence to the expression of defense genes related to ornamental sunflower.
