摘 要:目的研究蜂胶总黄酮(total flavonoids of propolis,TFP)的抗氧化活性及其对小鼠肝癌Hepa1-6细胞、人肺癌A549细胞和人胃癌SGC7901细胞增殖的抑制作用。方法通过测定TFP的总还原力及对1,1-二苯基-2-三硝基苯肼(DPPH)、羟基自由基和超氧阴离子自由基的清除能力,评价TFP的体外抗氧化活性;采用CCK-8法检测蜂胶总黄酮抗肿瘤细胞增殖的能力。结果实验结果表明,在25~150μg/mL浓度范围内TFP具有较强的总还原力,对DPPH自由基、羟基自由基和超氧阴离子自由基的清除能力均呈现浓度依赖性增大,最大清除率分别为(72.47±3.13)%、(70.84±4.01)%和(68.24±4.56)%;50、100、200μg/mL的TFP对Hepa1-6、A549和SGC7901细胞的生长均有明显的抑制作用,且在24~72 h时间范围内的抑制作用随时间延长而逐渐增大。结论TFP在体外具有较强的抗氧化能力,且对Hepa1-6、A549和SGC7901细胞的生长具有显著的抑制作用。Objective To study the antioxidant activity of total flavonoids extracted from propolis(TFP).The inhibitory effect of TFP on the proliferation of Hepa1-6,A549 and SGC7901 cells. Methods The antioxidant activity of TFP were evaluated by measuring the total reducing power and its scavenging ability to DPPH,hydroxyl and superoxide anion free radicals. Growth inhibitory effect of total flavonoids on tumor cells was assessed by CCK-8 assay. Results Results showed that the TFP had strong reducing ability,DPPH、hydroxyl and superoxide anion free radical scavenging rate could reach(72.47±3.13)%,(70.84±4.01)% and(68.24±4.56)%,respectively. The TFP at the concentration of 50,100,200 μg/mL significantly inhibited the growth of Hepa1-6,A549 and SGC7901 cells,and the inhibitory effect increased with time in the range of 24 to 72 hours. Conclusion TFP had strong antioxidant capacity in vitro and could significantly inhibit the proliferation of Hepa1-6,A549 and SGC7901 cells.
