摘 要:目的探究益肾通脉方对急性脑梗死缺血再灌注大鼠氧化应激、脑组织形态学改变及行为学的影响。方法选取180只健康SD雄性大鼠,通过随机分组的方式对大鼠进行分组,分为假手术组,模型组与低、中、高剂量益肾通脉方组,每组36只。给予益肾通脉方组大鼠中药灌胃(低剂量组:25 mg/kg;中剂量组:50 mg/kg;高剂量组:100 mg/kg),给予假手术组与模型组大鼠等体积生理盐水灌胃,连续1周。制备大脑中动脉闭塞后再灌注大鼠模型,筛选出建模成功的大鼠,每组32只。假手术组大鼠在建模过程中仅进行血管分离,不进行动脉栓塞。治疗14 d后,各组分别取8只大鼠断头取脑制备成匀浆,按照试剂盒的操作说明检测超氧化物歧化酶(SOD)、丙二醛(MDA)、NO的含量水平;各组分别取8只大鼠断头取脑制备成匀浆,用酶标仪检测ATP酶活力情况;分别于治疗7、14 d对各组大鼠神经行为学进行评估与记录;治疗14 d后,各组随机抽取8只大鼠,断头取脑制备成匀浆,通过比色法测定大鼠脑组织caspase-3、caspase-8、caspase-9活性情况;治疗14 d后,各组随机抽取8只大鼠,断头取脑制成切片并镜检,观察各组大鼠脑组织形态学改变情况;并通过显微图像分析系统计算梗死灶体积及梗死率。结果治疗14 d后,模型组大鼠SOD水平明显低于假手术组,MDA、NO水平明显高于假手术组(P<0.05);各剂量益肾通脉方组氧化应激指标与模型组比较差异有统计学意义(P<0.05)。模型组Na+-K+-ATP酶活力与Mg2+-ATP酶活力均明显低于假手术组(P<0.05);各剂量益肾通脉方组ATP酶活力均明显高于模型组(P<0.05)。治疗7、14 d之后,模型组大鼠神经行为学评分均明显高于假手术组(P<0.05);各剂量益肾通脉方组神经行为学评分均明显低于模型组(P<0.05)。治疗14 d后,模型组大鼠caspase-3、caspase-8、caspase-9表达水平明显强于假手术组(P<0.05);各剂量益肾通脉方组大鼠caspase-3�Objective To investigate the effects of Yishen Tongmai Decoction on the improvement of oxidative stress,brain tissue morphology and behavior in rats with acute cerebral infarction after ischemiareperfusion. Methods A total of 180 healthy SD male rats were randomly divided into sham operation group,model group and low,medium and high dose Yishen Tongmai group,with 36 rats in each group. Rats in Yishen Tongmai group were given traditional Chinese medicine by gavage(low dose group:25 mg/kg;medium dose group:50 mg/kg;high dose group:100 mg/kg),rats in the sham operation group and the model group were gavaged with normal saline of the same volume for 1 week. The rat model of reperfusion after middle cerebral artery occlusion was prepared and 32 rats in each group were screened out. During the modeling process,the rats in the sham operation group only underwent vascular separation,not arterial embolization. After 14 d of treatment,8 rats in each group were taken from their severed heads to prepare homogenate,and the levels of SOD,MDA and NO were detected according to the instructions of the kit. In each group,8 rats were cut off and their brains were extracted to prepare homogenate. ATP activity was detected by enzyme marker. The neurobehavioral behaviors of the rats in each group were evaluated and recorded on day 7 and day 14,respectively. After 14 d of treatment,8 rats were randomly selected from each group and their brains were cut off to prepare homogenate. The activity of caspase-3,caspase-8 and caspase-9 in the brain tissues of the rats was determined by colorimetry. After 14 days of treatment,8 rats were randomly selected from each group,and their brains were cut off and sectioned for microscopic examination. The infarct volume and the infarct rate were calculated by micrograph analysis system. Results After 14 d of treatment,SOD level in the model group was significantly lower than that in the sham group,and MDA and NO levels were significantly higher than that in the sham group(P<0.05).
