摘  要:目的:采用H2O2诱导的人黑素细胞体外氧化应激模型探讨紫草素对人黑素细胞的氧化应激损伤的干预作用研究。方法:在培养的黑素细胞加入250μmol/LH2O2作用12h,作为黑素细胞氧化应激模型,实验分为给药组、阳性对照组和空白组,分别选用紫草素、N-乙酰半胱氨酸(NAC)、20μL1640培养液进行干预。采用CCK8法检测黑素细胞活力,流式凋亡检测法检测黑素细胞凋亡率。结果:250μmol/LH2O2作用12h后的黑素细胞活力降低,凋亡增加,0.1μg/L、0.25μg/L、0.5μg/L的紫草素可明显增强细胞活力,降低黑素细胞凋亡率,与空白组比较有统计学意义(P<0.001),高浓度(1μg/L、2μg/L)的紫草素可降低黑素细胞活力,增加黑素细胞凋亡率。结论:紫草素在较低的浓度下(0.1μg/L^0.5μg/L),可增强细胞活力,降低黑素细胞凋亡率,其作用强度随浓度增高而增加。紫草素浓度大于1μg/L时,黑素细胞活力降低,凋亡率升高,表现为浓度越高,黑素细胞活力越低,凋亡率越高。Objective: To investigate the effect of shikonin on oxidative stress injury of human melanocytes by using an in vitro oxidative stress model induced by H2O2.Methods: The cultured melanocytes were treated with 250μmol/L H2O2 for 12 h as the model of oxidative stress of melanocytes.The experiment was divided into a drug administration group, a positive controlled group and a blank group.Shikonin, N-acetylcysteine(NAC) and 20μL1640 medium were used to intervene.The activity of melanocytes was detected by CCK8 and the apoptosis rate of melanocytes was detected by flow cytometry.Results: After 12 h treatment by 250μmol/L H2O2,the activity of melanocytes was decreased and the apoptosis was increased.Shikonin of 0.1μg/L,0.25μg/L and 0.5μg/L significantly increased the cell activity and decreased the apoptosis rate of melanocytes, which was statistically significant compared with the blank group(P < 0.001).High concentration of shikonin(1μg/L,2μg/L) decreased the activity of melanocytes and increased the apoptosis rate of melanocytes.Conclusion: Shikonin at low concentration(0.1~0.5μg/L) can enhance cell vitality and reduce the apoptosis rate of melanocytes, and its effect intensity increases with the increase of concentration.When the concentration of shikonin was more than 1μg/L,the activity of melanocytes decreased and the apoptosis rate increased.
