补肾活血方对精索静脉曲张大鼠附睾组织低氧诱导因子-1α及生育力的影响

摘　　要：目的:观察补肾活血方对精索静脉曲张大鼠附睾组织中低氧诱导因子-1α(HIF-1α)及生育力的影响,探讨其治疗VC性不育的可能机制。方法:将68只青春期雄性SD大鼠随机抽取8只为空白对照组,其余60只通过缩窄左肾静脉成功建立精索静脉曲张模型后,随机分为模型组,补肾活血方强精胶囊高、中、低剂量组,阳性对照组。分别给予生理盐水、强精胶囊混悬液、五子衍宗胶囊与少腹逐瘀胶囊混悬液灌胃,每天1次,连续4w。利用蛋白免疫印迹(Western Blot)和实时荧光定量-聚合酶链反应(RT-PCR)检测附睾组织中的HIF-1α的蛋白水平与mRNA水平的表达,并观察雄性大鼠精子密度与活力的变化及雌性大鼠受孕情况。结果:与空白对照组比较,模型组精子密度与活力显著降低(P<0.01),HIF-1α的蛋白表达明显升高和mRNA表达明显增强(P<0.01)。与模型组比较,强精胶囊高剂量组、中、低剂量组及阳性对照组的精子密度与活力明显提高(P<0.01),HIF-1α的蛋白表达减少和mRNA表达减弱(P<0.01)。与生育期雌性大鼠合笼后,发现强精胶囊高剂量组受孕率最高。结论:补肾活血方强精胶囊可以改善VC大鼠精子密度与活力,抑制附睾组织HIF-1α的蛋白水平和mRNA水平的过度表达,改善附睾组织低氧状态,从而促进附睾精子成熟,提高生育力。Objective:To observe the effect of Bushen Huoxue Recipe on hypoxia-inducible factor-1α(HIF-1α)and its fertility in epididymis for rats with varicocele,and to explore its possible mechanism of treatment of VC infertility.Methods:Eight out of 68 male Sprague-Dawley rats were randomly selected as the blank control group,and the remaining 60 successfully established varicocele model rats by narrowing the left renal vein were randomly divided into model group,Qiangjing capsule high,medium,and low dose group,positive controlled group.The saline,Qiangjing Capsule suspension,Wuzi Yanzong Capsule and Shaofuzhuyu Capsule suspension were administered to the stomach once a day for 4 weeks.The protein levels and mRNA levels of HIF-1α in epididymis were detected by Western Blot and real-time fluorescence quantitative polymerase chain reaction(RTPCR),and the changes of sperm density and vitality in male rats and the conception of pregnancy in female rats were observed.Results:Compared with the blank controlled group,the sperm density and vitality of the model group were significantly decreased(P<0.01),the protein expression of HIF-1α was significantly increased and the mRNA expression was significantly increased(P<0.01).Compared with the model group,the sperm density and vitality of the high,middle and low-dose groups of Qiangjing Capsule and the positive controlled group were significantly increased(P<0.01),and the protein and mRNA expression of HIF-1α was decreased(P<0.01).After co-cage with female rats in the growth period,it was found that the high-dose group of Qiangjing Capsule had the highest pregnancy rate.Conclusion:Bushen Huoxue Recipe can improve sperm density and vitality in varicocele rats,inhibit the over-expression of the protein and mRNA level of HIF-1α in epididym is,improve the hypoxic state of epididymis,and promote the maturation of epididymis sperm and improve fertility.
