摘 要:选择3种菊花提取物中杀虫效果最好的一种处理黄褐天幕毛虫幼虫(Malacosoma neustria testacean),测定其对黄褐天幕毛虫保护酶、解毒酶活力以及蛋白质和碳水化合物含量的影响,分析其对黄褐天幕毛虫幼虫营养状况及酶活的影响机制。采用超声提取的方法,获得三种菊花黑心菊(Rudbeckia hirta)、波斯菊(Cosmos bipinnata)、菊芋(Helianthus tuberosus)3种菊花提取物。在20 mg/mL浓度下,测定了三种菊花提取物对黄褐天幕毛虫的毒力,选择杀虫效果最好的菊芋提取物对天幕毛虫进行处理,测定对黄褐天幕毛虫12 h、24 h、48 h、72 h解毒酶GST和保护酶SOD、POD、CAT、PPO活力及蛋白质含量、碳水化合物含量及海藻糖含量。结果表明在20 mg/mL浓度下,菊芋花提取物对黄褐天幕毛虫的致死率高达97.52%。对黄褐天幕毛虫的保护酶POD、CAT主要表现为激活作用,而解毒酶GST活性明显被抑制;菊芋提取物诱导了蛋白质含量增加,碳水化合物和海藻糖含量则显著下降。20 mg/mL菊芋花提取物对黄褐天幕毛虫体内解毒酶和保护酶系统有干扰作用,对蛋白质含量有诱导促进作用。此外,菊芋花提取物还影响黄褐天幕毛虫体内碳水化合物的转化利用,使碳水化合物和海藻糖含量下降。Among the three kinds of chrysanthemum extracts,the larvae(malacosoma neustria testacean)were treated with the best insecticidal effect,and their effects on the protective enzyme,detoxification enzyme activity and protein and carbohydrate content were determined.Three kinds of chrysanthemum extracts were obtained by ultrasonic extraction,including chrysanthemum(Rudbeckia hirta),Persian chrysanthemum(cosmos bipinnata)and Jerusalem artichoke(helianthus tuberosus).The virulence of three chrysanthemum extracts to malacosoma neustria testacean was determined at 20 mg/ml concentration.the best insecticidal extract was selected for the treatment of malacosoma neustria testacean.The activity and protein content,carbohydrate content and trehalose content 12h、24h、48h、72h detoxifying enzyme GST and protective enzyme SOD、POD、CAT、PPO malacosoma neustria testacean.At 20 mg/ml concentration,the death rate of Jerusalem artichoke extract to malacosoma neustria testacea n was 97.52%.the protective enzyme POD、CAT for malacosoma neustria testacean mainly showed activation,while the activity GST detoxification enzyme was obviously inhibited;inulin extract induced an increase in protein content,while carbohydrate and trehalose content decreased significantly.At 20 mg/ml the extract of Jerusalem artichoke can interfere with detoxification enzyme and protective enzyme system of malacosoma neustria testacean protein content.In addition,the extract of Jerusalem artichoke also affected the conversion and utilization of carbohydrates in malacosoma neustria testacean,decreased the contents of carbohydrates and trehalose.a was decomposed under the action of inulin extract,which provided nutrition for insect growth and development.
