三物黄芩汤对白塞氏病小鼠炎症因子及STAT3蛋白的影响

摘　　要：目的:通过研究中药制剂三物黄芩汤对白塞氏病小鼠外周血中炎症因子IFN-γ及P-STAT3蛋白表达的影响,探讨三物黄芩汤的疗效机制。方法:BALB/c小鼠60只,分为空白组、模型组、沙利度胺组、三物黄芩汤组,每组10只。采用对虾原肌球蛋白过敏原,建立小鼠白塞氏病模型,采用ELISA方法检测小鼠外周血IFN-γ的水平变化,采用Western Blot检测小鼠脾脏相关蛋白P-STAT3含量。结果:与空白组小鼠比较,模型组的炎症因子水平及P-STAT3蛋白含量明显升高(P<0.01),表明白塞氏病小鼠的炎症因子水平及P-STAT3蛋白含量显著升高。给予沙利度胺、三物黄芩汤后,与模型组相比小鼠的炎症因子水平及P-STAT3蛋白含量明显降低(P<0.01或P<0.05),但沙利度胺组与中药高剂量组之间小鼠的炎症因子及P-STAT3蛋白含量水平无显著差异(P>0.05)。结论:三物黄芩汤可降低白塞氏病小鼠炎症因子水平及P-STAT3蛋白的表达。Objective:To study the therapeutic effect of Sanwu Huangqin Decoction on the protein expressions of inflammatory factors IFN-γand P-STAT3,in peripheral blood of mice with Behcet’s disease,thus to explore the action mechanism of Sanwu Huangqin Decoction.Methods:60 BALB/c mice were randomly divided into the blank group,the model group,Thalidomide group and Sanwu Huangqin Decoction group,with 10 mice in each group.The mice model of Behcet’s disease was established by using prawn tropomyosin allergen.The level of IFN-γ in peripheral blood of mice was detected by ELISA method,and the content of spleen-related protein P-STAT3 in mice was detected by Western Blot.Results:Compared to those in the blank group,the levels of inflammatory factors and the content of P-STAT3 protein increased significantly in the model group(P<0.01),indicating that the inflammatory factor level and P-STAT3 protein content increased significantly in the mice with the Behcet’s disease.The levels of inflammatory factors and the content of P-STAT3 protein were significantly lower in Thalidomide group and Sanwu Huangqin Decoction group than those in the model group(P<0.01,P<0.05);of which there were no statistical differences between Thalidomide group and the high-dose group of Sanhu Huangqin Decoction group(P>0.05).Conclusion:Sanwu Huangqin Decoction can reduce the levels of inflammatory factors and the expression of P-STAT3 protein in mice with Behcet’s disease.
