中药贴膜对兔膝骨性关节炎关节液中IL-1、IL-6、THF-α、HA含量水平的影响

摘　　要：目的:通过研究中药贴膜治疗早期兔膝关节骨性关节炎模型关节液白细胞介素-1(IL-1)、白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)、透明质酸(HA)含量变化,评价其治疗膝骨性关节炎的效果。方法:应用Hulht方法建立兔右膝关节骨性关节炎模型,随机分为模型对照组、治疗1组、治疗2组、治疗3组。分别采用医用橡皮膏外敷、传统方法粉碎的中药粉外敷、玻璃酸钠关节内注射、中药贴膜外敷治疗,另取10只健康白兔作为正常组。治疗6周后采用Elisa法测定各组实验动物右膝关节液IL-1、IL-6、TNF-α的含量;采用放射免疫分析法测定各组实验动物右膝关节液HA的含量变化。结果:关节液中IL-1、IL-6、TNF-α的含量测定结果显示,正常组最低,模型对照组最高;治疗1、2、3组治疗后均有改善,其中治疗2、3组治疗结果优于治疗1组;治疗3组IL-1、IL-6检测结果与治疗2组无差别,TNF-α含量检测结果低于治疗2组。关节液中HA含量测定结果显示模型对照组最低;治疗1、2、3组治疗后均优于模型对照组,其中治疗2组最高,治疗3组其次,治疗1组最低。结论:中药贴膜可以改善动物模型关节液中IL-1、IL-6、TNF-α、HA含量水平,推测中药贴膜治疗骨性关节炎作用机理与降低关节液中IL-1、IL-6、TNF-α,增加HA含量有关。Objective: To evaluate the effect of traditional Chinese medicine patch on the changes of interleukin-1(IL-1), interleukin-6(IL-6), tumor necrosis factor-α(TNF-α) and hyaluronic acid(HA) in articular fluid the treatment of early stage of knee osteoarthritis(KOA) in rabbits. Methods: A model of right knee osteoarthritis was established by Hulht method in rabbits, the successful models were randomly divided into the model control group(medical plaster), the treatment group 1(traditional Chinese medicine powder), the treatment group 2(sodium hyaluronate injection) and the treatment group 3(traditional Chinese medicine patch). Another 10 healthy white rabbits were taken as the normal control. After 6 weeks of the treatment, the Elisa method was used to determine the contents of IL-1, IL-6 and TNF-α of right knee joint fluid, and radioimmunoassay(RIA) was used to determine the HA content of right knee joint fluid in the groups. Results: In terms of the contents of IL-1, IL-6 and TNF-α in joint fluid, they were the lowest in the normal control group, and they were the highest in the model control group;after the treatment, they were all improved in the treatment group 1, the treatment group 2 and the treatment group 3, and the treatment group 2 and the treatment group 3 were better than the treatment group 1;there were no statistical differences in the levels of IL-1 and IL-6 between the treatment group 3 and the treatment group 2, but the content of TNF-α was lower in the treatment group 3 than that in the treatment group 2. In terms of the content of HA in joint fluid, it was the lowest in the model control group;it was improved after the treatment in the treatment groups, among which the treatment group 2 has the best results, followed by the treatment group 3 and the treatment group 1. Conclusion: Traditional Chinese medicine patch can improve the levels of IL-1, IL-6, TNF-α and HA in joint fluids of the animal model.
