摘 要:目的考察3种提取方法对二黄汤中甘草苷、黄芩苷、巴马汀、小檗碱、甘草次酸在大鼠体内药动学的影响。方法分别采用半仿生-酶法、半仿生法、醇提法提取二黄汤。大鼠灌胃给予提取液(≤1000 Da,15.0 g/kg)后,于0.083、0.167、0.333、0.5、1、2、3、4、6、8、12、24、48 h采血,HPLC-MS/MS法测定5种成分血药浓度,计算主要药动学参数。结果半仿生提取液中甘草苷tmax更短,Cmax、AUC0~t更高;半仿生-酶提取液中黄芩苷、巴马汀、小檗碱、甘草次酸Cmax、AUC0~t更高,其中巴马汀、小檗碱tmax更短。结论半仿生-酶法、半仿生法有利于二黄汤中5种成分的提取,可提高其口服吸收体内生物利用度。AIM To investigate the effects of three extraction methods on the pharmacokinetics of liquiritin,baicalin,palmatine,berberine and glycyrrhizic acid in Erhuang Decoction in rats in vivo.METHODS(Semi-bionic)-enzyme method,semi-bionic method and ethanol extraction method were adopted in the extraction for Erhuang Decoction,respectively.Rats were given intragastrical administration of extractions(≤1000 Da,15.0 g/kg),after which blood collection was made at 0.083,0.167,0.333,0.5,1,2,3,4,6,8,12,24,48 h,HPLC-MS/MS was adopted in the plasma concentration determination of five constituents,and main pharmacokinetic parameters were calculated.RESULTS In the semi-bionic extract,liquiritin demonstrated shorter tmax and higher Cmax,AUC0-t.In the(semi-bionic)-enzyme extract,baicalin,palmatine,berberine and glycyrrhizic acid displayed higher Cmax and AUC0-t,among which palmatine and berberine exhibited shorter tmax.CONCLUSION(Semi-bionic)-enzyme method and semi-bionic method are beneficial to the extraction for five constituents in Erhuang Decoction and can enhance their in vivo bioavailabilities of oral administration.
