摘 要:目的建立UHPLC-PDA法同时测定毛大丁草Gerbera piloselloides(Linn.)Cass.中熊果苷、绿原酸、木犀草苷、异绿原酸B、异绿原酸A、异绿原酸C的含量。方法毛大丁草50%甲醇提取物的分析采用Waters BEH C18色谱柱(2.1 mm×150 mm,1.7μm);流动相乙腈-0.1%甲酸,梯度洗脱;体积流量0.28 mL/min;柱温40℃;检测波长280、328 nm。结果熊果苷、绿原酸、木犀草苷、异绿原酸B、异绿原酸A、异绿原酸C分别在4.45~284.59(r=0.9999)、0.30~19.52(r=0.9998)、0.30~19.50(r=0.9998)、0.31~19.84(r=0.9999)、0.61~38.96(r=0.9998)、0.32~20.19(r=0.9998)μg/mL范围内线性关系良好,平均加样回收率分别为102.06%、103.37%、104.97%、104.79%、105.21%、104.75%,RSD分别为1.88%、1.70%、1.20%、1.89%、1.53%、1.25%。结论该方法稳定,重复性好,可用于毛大丁草的质量控制。AIM To establish a UHPLC analysis method for simultaneous determination of arbutin,chlorogenic acid,luteoloside,isochlorogenic acid B,isochlorogenic acid A and isochlorognic acid C in Gerbera piloselloides(Linn.)Cass..METHODS The analysis of 50%methanol extract of G.piloselloides was performed on a 40℃thermostatic Waters BEH C18 column(2.1 mm×150 mm,1.7μm),with the mobile phase comprising of acetonitrile and 0.1%formic acid flowing at 0.28 mL/min in a gradient elution manner,and the detection wavelengths were set at 280,328 nm.RESULTS Arbutin,chlorogenic acid,luteoloside,isochlorogenic acid B,isochlorogenic acid A and isochlorognic acid C showed good linear relationships within the ranges of 4.45-284.59(r=0.9999),0.30-19.52(r=0.9998),0.30-19.50(r=0.9998),0.31-19.84(r=0.9999),0.61-38.96(r=0.9998),0.32-20.19(r=0.9998)μg/mL,whose average recoveries were 102.06%,103.37%,104.97%,104.79%,105.21%,104.75%with the RSDs of 1.88%,1.70%,1.20%,1.89%,1.53%,1.25%,respectively.CONCLUSION This accurate,stable and reproducible method can be used for the quality control of G.piloselloides.
