摘 要:目的探究甲状腺乳头状癌(PTC)患者癌组织中长链非编码RNA(lncRNA)MIR31HG的表达变化及意义。方法选取PTC患者86例,术中采集其PTC组织及癌旁正常组织(距离肿瘤边缘>2 cm),用荧光定量PCR法检测组织中lncRNA MIR31HG表达,并分析其表达变化与患者临床病理特征的关系;对患者进行随访,统计5年存活率;用Kaplan-Meier生存曲线(Log-rank检验)分析lncRNA MIR31HG表达与PTC患者预后的关系;COX回归分析PTC患者不良预后的危险因素。结果PTC组织中lncRNA MIR31HG相对表达量较癌旁正常组织高(t=24.008,P<0.05)。不同性别、年龄患者PTC组织中lncRNA MIR31HG的表达比较差异无统计学意义(P均>0.05)。TNM分期Ⅲ、Ⅳ期,肿瘤直径>2 cm、局部浸润及伴淋巴结转移的患者PTC组织中lncRNA MIR31HG表达高于TNM分期Ⅰ、Ⅱ期,肿瘤直径≤2 cm、无局部浸润及无淋巴结转移的患者(P均<0.05)。以PTC组织中lncRNA MIR31HG表达的平均数1.468为界值,分为高表达组42例,低表达组44例。高表达组5年OS低于低表达组(χ2=4.949,P<0.05)。lncRNA MIR31HG高表达、肿瘤TNM分期高及淋巴结转移是PTC患者预后不良的独立危险因素。结论lncRNA MIR31HG在PTC组织中呈高表达,其可能参与肿瘤的发生发展,并影响患者预后。Objective To explore the expression and significance of long non-coding RNA(lncRNA)MIR31HG in the papillary thyroid carcinoma(PTC).Methods Eighty-six patients with PTC were selected.Their PTC tissues and adjacent normal tissues(more than 2 cm away from the tumor edge)were collected during the operation.The expression of lncRNA MIR31HG in the tissues was detected by fluorescence quantitative PCR,and the relationship between the expression changes and clinicopathological characteristics of the patients was analyzed.The patients were followed up and the 5-year survival rate was recorded;the relationship between lncRNA MIR31HG expression and prognosis of PTC patients was analyzed by Kaplan-Meier survival curve(Log-rank test);the risk factors of poor prognosis of PTC patients were analyzed by Cox regression.Results The relative expression of lncRNA MIR31HG in PTC cancer tissues was higher than that in the adjacent normal tissues(t=24.008,P<0.05).There was no significant difference in the expression of lncRNA MIR31HG in PTC tissues of different gender,age(all P>0.05).The expression of lncRNA MIR31HG in PTC patients with TNM stage III and IV,tumor diameter>2 cm,local invasion and lymph node metastasis was higher than that in patients with TNM stage I and II,tumor diameter≤2 cm,no local invasion,and no lymph node metastasis(all P<0.05).The patients were divided into the high expression group of 42 cases and low expression group of 44 cases by taking the average lncRNA MIR31HG expression 1.468 in PTC cancer tissues as the cut-off value.The 5-year OS of the high expression group was lower than that of low expression group(χ2=4.949,P<0.05).High expression of lncRNA MIR31HG,high TNM stage,and lymph node metastasis were independent risk factors for poor prognosis of PTC patients.Conclusion LncRNA MIR31HG is highly expressed in PTC tissues,which may be involved in the development of tumor and affect the prognosis of patients.
