养阴温阳方对C57 BL/6小鼠肿瘤生长及p53/GADD45A/BIM信号通路的影响

摘　　要：目的观察养阴温阳方抗肺癌的可能作用机制。方法建立Lewis肺癌C57 BL/6皮下移植瘤模型,随机分为4组:对照组,顺铂组,养阴温阳方组,顺铂+养阴温阳方组,每组8只。对照组每日给予0.2 m L 0.9%氯化钠溶液灌胃,顺铂组每4 d给予2 mg/kg腹腔注射,养阴温阳方组每日给予11.33 g/kg养阴温阳方灌胃,顺铂+养阴温阳方组每日给予11.33 g/kg养阴温阳方灌胃,顺铂使用方案同顺铂组,共计14 d。观察各组肿瘤体积、瘤体质量,计算抑瘤率;HE染色观察各组肿瘤病理变化,免疫组化检测突变型p53、GADD45A、BIM蛋白表达情况。结果与对照组相比,顺铂组、养阴温阳方组、顺铂+养阴温阳方组均可明显抑制肿瘤生长(P <0.05),顺铂+养阴温阳方组效果更好。HE染色显示,与对照组相比,顺铂组肿瘤组织细胞明显坏死,养阴温阳方组、顺铂+养阴温阳方组肿瘤组织明显出现炎性坏死。免疫组化法显示,养阴温阳方组突变型p53明显下降,GADD45A、BIM蛋白表达增加。结论养阴温阳方可明显抑制肿瘤生长,与顺铂联合可提高疗效,其机制可能与p53/GADD45A/BIM信号通路有关。Objective To observe the effect of Yangyin Wenyang Formula( YYWY) on tumor growth and P53/GADD45 A/BIM signaling pathway in Lewis lung cancer of C57 BL/6 mice.Methods The Lewis lung cancer C57 BL/6 subcutaneous xenograft model was established and the mice were randomly divided into 4 groups: control group,cisplatin group,YYWY group,and cisplatin combined with YYWY group.The mice were sacrificed on the 14 th day.The inhibition rate,tumor volume and tumor weight of each group were observed.HE staining was used to detect the pathological changes.Immunohistochemistry of each group was used to detect the expressions of p53,GADD45 A and BIM protein.Results Compared with the control group,cisplatin group,YYWY group and combination group could significantly inhibit tumor growth( P < 0.05),and the combination group was better.The results of HE staining showed that the tumor tissue showed obvious necrosis after administration,and the results of immunohistochemistry showed that YYWY could reduce the expressions of mutant p53,GADD45 A and BIM protein.Conclusion YYWY can significantly inhibit tumor growth,and combined with cisplatin can improve the efficacy,and its mechanism may be related to p53/GADD45 A/BIM signaling pathway.
