温肾健脾膏对疲劳综合征大鼠免疫机制的影响

摘　　要：目的探讨温肾健脾膏对脾肾阳虚型疲劳综合征大鼠免疫机制的影响。方法采用SPF级健康雄性SD大鼠90只,随机均分为空白组、模型组、中药组,每组30只。将模型组、中药组用于脾肾阳虚型疲劳综合征大鼠模型制备(束缚+冷水游泳刺激法),中药组采用温肾健脾膏[1.8 g/(kg·d)]治疗,模型组与空白组灌胃等体积生理盐水,连续治疗60 d。观察各组大鼠肝功能、肾功能情况,流式细胞法检测各组大鼠外周血T淋巴细胞亚群分布情况,MTT法检测脾脏T、B淋巴细胞增殖能力,酶联免疫吸附(ELISA)法检测外周血清炎性因子水平。结果与空白组比较,模型组的外周血清ALT、AST、BUN、Scr、CD+8、IL-6、IL-2水平明显升高,CD+3、CD+4、CD+4/CD+8、IL-4、IFN-γ明显降低,差异有统计学意义(P <0.05);与模型组相比,中药组的外周血清ALT、AST、BUN、Scr、CD+8、IL-6、IL-2水平明显降低,CD+3、CD+4、CD+4/CD+8、IL-4、IFN-γ明显升高,差异显著,有统计学意义(P <0.05);脾脏T、B淋巴细胞增殖能力结果显示,模型组T、B淋巴细胞增殖明显高于空白组(P <0.05),中药组T、B淋巴细胞增殖明显低于模型组(P <0.05)。结论温肾健脾膏对脾肾阳虚型疲劳综合征大鼠的肝肾功能有改善作用,可能与调节T淋巴细胞亚群分布、T、B淋巴细胞增殖能力有关。Objective To explore the effects of Wenshen Jianpi Ointment on immune mechanism in rats with fatigue syndrome( FS).Methods Ninety SPF-level healthy male SD rats were randomly divided into blank group,model group and TCM( traditional Chinese medicine) group,30 cases in each group.In model group and TCM group,the rates with FS of spleen-kidney Yang deficiency were prepared( bondage combined with swimming stimulation in cold water).TCM group was treated with Wenshen Jianpi Ointment[1.8 g/( kg·d) ],while model group and blank group were intragastrically administered with the same volume of normal saline for 60 d.The liver and kidney function situations in each group were observed.The distributions of T lymphocyte subsets in peripheral blood in each group were detected by flow cytometry.The proliferation ability of T and B lymphocytes in spleen was tested by MTT.The levels of serum inflammatory factors in peripheral blood were detected by enzyme-linked immunosorbent assay( ELISA).Results Compared with those of the blank group,serum levels of ALT,AST,BUN,Scr,CD8+,IL-6 and IL-2 in peripheral blood were significantly increased,while levels of CD^3+,CD^4+,CD4+/CD^8+,IL-4 and IFN-γwere significantly decreased in the model group( P < 0.05).Compared with those of the model group,serum levels of ALT,AST,BUN,Scr,CD8+,IL-6 and IL-2 in peripheral blood were significantly decreased,while levels of CD^3+,CD^4+,CD^4+/CD^8+,IL-4 and IFN-γ were significantly increased in TCM group( P < 0.05).The proliferation abilities of T and B lymphocytes in spleen in the model group were significantly higher than those in the blank group( P < 0.05),which were significantly lower in TCM group than those in the model group( P < 0.05).Conclusion Wenshen Jianpi Ointment can improve liver and kidney function of rats with FS of spleen-kidney Yang deficiency,which may be related to regulating distributions of T lymphocyte subsets and proliferation abilities of T and B lymphocytes.
