摘  要:目的探讨土家族麝针疗法对缺血性脑卒中大鼠神经功能的康复作用及对下丘脑中神经递质多巴胺(DA)、5-羟色胺(HT)和炎性因子细胞间黏附分子(ICAM)-1、单核细胞趋化蛋白(MCP)-1含量变化的影响。方法采用改良线栓法建立缺血性脑卒中大鼠模型,取造模成功的大鼠,随机分为模型组、土家族麝针组和针刺组,另取大鼠不行线栓栓塞作为假手术组,每组30只。麝针组和针刺组分别以自制麝针和普通毫针刺激大鼠对侧头皮运动区肌层,治疗3个疗程;假手术组和模型组不做任何处理。分别于治疗前及各疗程治疗结束后取各组大鼠以平衡木法评定神经运动功能,采用酶联免疫吸附试验(ELISA)检测下丘脑中DA、5-HT、ICAM-1和MCP-1的含量。结果与假手术组比较,治疗前,缺血性脑卒中模型大鼠神经运动功能评分显著升高,下丘脑组织中DA、ICAM-1和MCP-1含量明显升高,5-HT含量显著降低;经过3个疗程治疗后,相对于模型组及针刺组,麝针组大鼠的运动功能恢复更加接近于假手术组,土家族麝针疗法降低下丘脑组织中DA、ICAM-1和MCP-1及提高5-HT含量作用更加明显。结论土家族麝针疗法能显著促进缺血性脑卒中模型大鼠神经运动功能的恢复,其作用机制可能与其降低脑组织炎性因子MCP-1、ICAM-1含量,抑制神经递质DA释放和促进5-HT的释放有关。Objective To investigate the effect of Tujia musk acupuncture on the content of DA,5-HT,ICAM-1 and MCP-1 in the rats with ischemic stroke and explore the mechanism of Tujia musk acupuncture in treating ischemic stroke.Methods Modified thread-occlusion method was used to establish the ischemic stroke rat model.The successful model rats were randomly divided into model group,Tujia musk acupuncture group and the acupunctured group and normal rats without thread-occlusion were used as sham group,30 rats in each group.Rats in Tujia musk acupuncture group and acupunctured group were treated by the homemade acupuncture needle contained musk and filiform needle to stimulate the muscular layer of the lateral scalp respectively for three courses.There was no operation for model group and sham group.After the treatment,the bar walking test was used to evaluate the motor function of each group,ELISA method was used to detect the contents of DA,5-HT,ICAM-1 and MCP-1 in the hypothalamus of the rats.Results Before treatment,compared with sham group,the motor function decreased significantly,the contents of DA,ICAM-1 and MCP-1 reduced significantly while the contents of 5-HT increased significantly in model group.After three courses of treatment,compared with model group and acupuncture group,the motor function of the rats in Tujia acupuncture group recovered significantly and was close to that of sham group.Tujia acupuncture treatment showed more obvious effect.Conclusions Tujia musk acupuncture could improve the motor function of rats with ischemic stroke,which might be related to reducing the content of DA,ICAM-1 and MCP-1 while increasing the content of 5-HT in the hypothalamic.
