摘 要:目的比较低共熔溶剂与乙醇用于制备黄芩总黄酮提取物的差异。方法分别以氯化胆碱-乳酸(摩尔比1∶2)或65%乙醇作为溶剂提取黄芩药材,所得提取液与水混合得混悬液,并进一步经冷冻干燥制得提取物粉末,观察并比较两种提取物的外观形态、粒径、溶解度和体外溶出度。结果低共熔溶剂和乙醇提取液中总黄酮含量分别为(12.64±0.86) mg·g-1和(0.19±0.07) mg·g-1,相应的冻干粉中总黄酮含量分别为(8.97±0.55)mg·g-1和(0.69±0.07)mg·g-1,且混悬液及冻干粉复溶后粒径均小于1μm。两种冻干粉中总黄酮溶解度均高于黄芩苷原料药(P均<0.05),且在水中45 min时累积溶出即达90%以上,而黄芩苷原料药2 h时溶出度不足30%。结论低共熔溶剂对黄芩总黄酮提取效率优于乙醇,且能改善所得提取物溶解和溶出性能,有利于进一步制剂。Objective To compare the performance of deep eutectic solvent(DES)and ethanol on preparation of total flavonoids extract from Scutellaria baicalensis Georgi. Methods Scutellaria baicalensis was extracted by ethanol or DES(choline chloride and lactic acid at molar ratio of 1∶2). The extraction liquids were mixed with water to obtain suspensions,and followed by freeze-drying to prepare extract powders. The extracts were compared on their appearance,particle size,and saturated solubility and in vitro dissolution profiles in water.Results Content of total flavonoids in extraction liquid was(12.64±0.86)mg·g-1 for DES and(0.19±0.07)mg·g-1 for ethanol,and that was(8.97±0.55)mg·g-1 and(0.69±0.07)mg·g-1 for DES and ethanol extract powders,respectively. The extract powder of both DES and ethanol showed a larger solubility in water than baicailin itself(P all<0.05). And their dissolution rates reached over 90% at 45 min,whereas baicailin released less than 30% at 2 h. Conclusion DES showed a better extraction efficiency than ethanol,and the obtained extract possessed favorable solubility and dissolution properties,which is advantageous for further preparation process.
