摘 要:为明确新疆栽培紫花苜蓿(Medicago sativa)矮化、丛枝等疑似病毒病症状的病原,本研究应用小RNA测序结合RT-PCR技术开展了病原分子检测与鉴定。结果表明,在36份样品中扩增得到一条长约1000 bp的特异性片段,该片段序列与已报道的其他苜蓿矮化病毒分离物聚合酶蛋白(L)基因的一致性为84.50%~88.24%。构建的系统发育树表明该病毒为苜蓿矮化病毒(Alfalfa dwarf virus),隶属于弹状病毒科(Rhabdoviridae)细胞质弹状病毒属(Cytorhabdovirus)。首次报道了苜蓿矮化病毒病在中国的发生。In recent years,it has been found that cultivated alfalfa(Medicago sativa)has symptoms of suspected virus disease such as dwarf,witches’broom in Xinjiang.In order to determine its pathogen,this study applied small RNA sequencing technology combined with RT-PCR technology to carry out pathogen molecular detection and identification.The results showed that a specific fragment of about 1000 bp was amplified in 36 samples,and the sequence of this fragment was 84.50%-88.24%identical to the polymerase protein(L)gene of other Alfalfa dwarf virus(ADV)isolates reported.The constructed phylogenetic tree confirmed that the virus belonged to the Rhabdoviridae Cytorhabdovirus.The results of this study are the first to report the occurrence of Alfalfa dwarf virus disease in China.
