摘 要:探讨在普通牛乳中添加"等同母乳剂量"的生长抑素(SST)及胃动素(MTL),对牛乳过敏症的预防调节作用及其对自身胃肠肽分泌的影响。新生BN大鼠,给与3种喂养方式加皮肤涂抹致敏,喂养4周:纯母乳组、纯牛乳组、添加SST及MTL的牛乳喂养组(SST+MTL组),每周量化评定"临床损害"评分。酶联免疫法(ELISA)测定血总免疫球蛋白E(IgE)水平、大便及血中的SST及MTL浓度。动物处死前测定胃排空比率及小肠推进比。结果母乳组、牛乳组及SST+MTL组的各项值分别为:IgE (ng/mL) 45.75±5.05,580.42±45.24,209.49±17.59;临床损害总评分为0.50±0.61,9.37±1.04,5.37±1.19;牛乳组IgE水平及临床损害评分均显著高于另两组(P<0.0001)。牛乳组的胃动力较母乳组显著低下,有明显小肠激惹及腹泻。SST+MTL组的胃动力较牛乳组有明显改善,腹泻症状轻症化,胃肠动力状况更接近母乳组。牛乳组大便及血中SST/MTL水平较母乳组均有显著低下,以大便中水平下降更为显著。SST+MTL组的大便中SST/MTL浓度较牛乳组提升约3倍,而血中SST/MTL浓度仅提升10%~20%。SST+MTL组的大便及血中SST/MTL水平更接近母乳组状况。结果表明普通牛乳中添加了SST及MTL后,能减轻牛乳的致过敏性,能提升其胃肠道耐受性,且SST与MTL在胃肠动力调节上有拮抗/平衡作用,而对幼崽血循环中胃肠肽水平的影响则相对较小。Somatostatin(SST)and motilin(MTL)are abundantly expressed in human breast milk.In this study,the same dose of SST and MTL as breast milk were added into cow milk,and its regulatory effects on food allergy and gastrointestinal peptide secretion were explored.New born Brown Norway rats were divided into three groups and given different feeding and skin smear sensitization for 4 weeks.The three groups were fed with pure breast milk,pure cow milk,cow milk added SST and MTL(SST+MTL group).Clinical damages were evaluated and quantified into scores level every week.Levels of serum total IgE,SST and MTL in fecal and blood were measured by enzyme linked immune sorbent assay.The gastric emptying ratio and intestinal propulsive ratio were measured before rats executed.The detection values in breast milk group,cow milk group and SST+MTL group were respectively listed:IgE(ng/mL)45.75±5.05,580.42±45.24,209.49±17.59;score values of clinical damage 0.50±0.61,9.37±1.04,5.37±1.19.IgE level and clinical damage score in cow milk group were significantly higher than those of other two groups(P<0.0001).Gastric motility of cow milk group was significantly lower than breast milk group.Cow milk group have obvious symptoms of diarrhea,irritability and fast peristalsis of intestine.SST+MTL group was significantly improved on gastric motility and intestinal irritability comparing to cow milk group.Gastrointestinal motility of SST+MTL group was more similar to that of breast milk group.SST/MTL level of stool and blood in cow milk group were significantly lower than those of breast milk group,especially the stool level was more significantdecreased.SST/MTL levels of stool in SST+MTL group was about three times higher than that of milk group,while the blood levels were only increased about 10%~20%.The of SST/MTL levels of stool and blood in SST+MTL group were closer to that of breast milk group.Cow milk added SST and MTL helped to reduce the allergic reaction and improve gastrointestinal tolerance.SST and MTL had an antagonistic and bala
