摘  要:建立了小鼠肝脏中UV-327和UV-328两种苯并三唑类紫外线吸收剂的高效液相色谱测定方法。样品经正己烷-丙酮(体积比1:1)提取,Waters Symmetry■C18柱(250 mm×4.6 mm,5μm)分离,以甲醇为流动相,检测波长为340 nm。紫外线吸收剂UV-327和UV-328均在0.05~20.0μg/mL范围内线性关系良好(r≥0.9998),检出限和定量限分别为0.01μg/mL和0.03μg/mL。UV-327和UV-328低、中、高3种浓度(0.20、2.00、16.0μg/mL)加标样品的回收率为94.9%~98.5%与97.0%~102.2%;日内精密度为3.7%~5.6%与3.8%~5.3%;日间精密度为2.8%~8.2%与2.8%~9.5%;提取回收率为94.2%~98.9%与94.3%~97.8%;基质效应为92.5%~110.8%与92.3%~112.3%。该方法准确、简便,适用于小鼠肝中紫外线吸收剂UV-327和UV-328的测定。A high performance liquid chromatography method was developed and validated to determine two benzotriazole ultraviolet absorbents(UV-327 and UV-328)in mouse liver.The sample was extracted by hexane-acetone(1∶1,V/V)and then separated on a Waters Symmetry■ C18 column(250 mm×4.6 mm,5μm).The mobile phase was methanol and the detection wavelength was set at 340 nm.The calibration curves of UV-327 and UV-328 were linear(r≥0.9998)over the concentration range of 0.05-20.0μg/mL,and the limit of detection and the limit of quantitation were 0.01μg/mL and 0.03μg/mL,respectively.The recoveries of UV-327 and UV-328 at three different concentrations(0.20,2.00,16.0μg/mL)were from 94.9%to 98.5%and 97.0%to 102.2%.The intra-day precisions(n=6)of UV-327 and UV-328 were from 3.7%to 5.6%and 3.8%to 5.3%,while the inter-day precisions(n=3)were from 2.8%to 8.2%and 2.8%to 9.5%.The extraction recoveries of UV-327 and UV-328 were from 94.2%to 98.9%and 94.3%to 97.8%,and the matrix effects were from 92.5%to 110.8%and 92.3%to 112.3%.The method was accurate and simple,and suitable for the determination of UV-327 and UV-328 in mouse liver.
