摘 要:目的分析雷公藤多苷通过Akt/mTOR信号通路对糖尿病肾病(DN)小鼠肾损伤影响。方法选取SPF级C57 BL/6 J小鼠75只,按随机数字表法将小鼠分成5组,分别为对照组、模型组、雷公藤多苷低剂量组、雷公藤多苷中剂量组和雷公藤多苷高剂量组,除对照组外,其他4组小鼠制备DN模型,雷公藤多苷低、中、高剂量组分别灌胃剂量为25 mg/kg、50 mg/kg和100 mg/kg的雷公藤多苷悬浊液(10 mg雷公藤多苷溶于1 ml蒸馏水内,配置浓度是10 g/L),对照组和模型组灌胃等剂量0.9%氯化钠溶液,5组小鼠均连续灌胃8周。观察5组小鼠一般状况、体重、相对肾重、血清生化指标及肾脏组织mTOR、p-mTOR、Akt、p-Akt蛋白与mRNA表达量。结果模型组小鼠体重较对照组降低,左肾重、相对肾重较对照组升高(P<0.05);雷公藤多苷低、中、高剂量组小鼠体重较模型组升高,左肾重、相对肾重较模型组降低,差异有统计学意义(P<0.05);模型组小鼠血清TC、TG、LDL含量较对照组升高,HDL含量较对照组降低(P<0.05);雷公藤多苷低、中、高剂量组小鼠血清TC、TG、LDL含量较模型组降低,HDL含量较模型组升高,差异有统计学意义(P<0.05);模型组小鼠血清糖化血清蛋白、尿素氮、肌酐、FBG含量及总尿蛋白较对照组升高(P<0.05);雷公藤多苷低、中、高剂量组小鼠血清蛋白、尿素氮、肌酐、FBG含量及总尿蛋白较模型组降低,差异有统计学意义(P<0.05);模型组小鼠肾脏组织p-mTOR、p-Akt蛋白与mRNA表达较对照组升高(P<0.05);雷公藤多苷低、中、高剂量组小鼠肾脏组织p-mTOR、p-Akt蛋白与mRNA表达较模型组降低,差异有统计学意义(P<0.05)。结论雷公藤多苷可有效改善DN小鼠肾损伤情况,其作用机制可能和抑制Akt/mTOR信号通路活化有关。Objective To investigate the effects of tripterygium glycosides on the renal injury in mice with diabetic nephropathy via Akt/mTOR signaling pathway.Methods Seventy-five SPF C57 BL/6J mice were randomly divided into five groups:control group,model group,low dose tripterygium glycosides group,medium dose tripterygium glycosides group and high dose tripterygium glycosides group.The DN models were established in mice except for the mice in control group.The mice in low-dose,medium-dose and high-does groups of tripterygium glycosides were given tripterygium wilfordii turbid liquid by gavage,with 25mg/kg,50mg/kg,100mg/kg,respecyively(tripterygium glycosides 10mg dissolved in 1ml of distilled water at a concentration of 10g/L).The mice in control group and model group were intragastrically administered with isodose 0.9%sodium chloride solution.The mice in the five groups were continuously administered for 8 weeks.The general condition,body weight,relative kidney weight,serum biochemical parameters,the protein expressions and mRNA expressions of mTOR,pmTOR,Akt,pAkt in kidney tissues were observed and compared among the five groups.Results The body weight of the mice in model group was significantly lower than that in control group(P<0.05).The weight of the left kidney and the relative kidney weight in model group were significantly higher than those in control group(P<0.05).The weight of mice in low-dose,medium-dose and high-dose tripterygium glycosides treatement group was significantly higher than that in model group(P<0.05).The left kidney weight and the relative kidney weight of mice in low-dose,medium-dose and high-dose tripterygium glycosides treatement group were significantly lower than those in model group(P<0.05).Moreover the serum levels of TC,TG and LDL in model group were significantly higher than those in control group(P<0.05),however the HDL levels in model group were significantly lower than those in control group(P<0.05).The serum levels of TC,TG and LDL in low-dose,medium-dose and high-dose tripterygiu
