摘 要:目的研究子宫肌瘤患者血清miR-21表达水平,分析其与雌激素受体(ER)和孕激素受体(PR)相关性。方法子宫肌瘤患者118例作为子宫肌瘤组,另选择同期因子宫脱垂行子宫切除术患者118例作为正常子宫肌层组。实时荧光定量(qRT-PCR)法检测血清中miR-21、ER mRNA、PR mRNA表达水平,免疫组织化学染色检测组织中ER、PR蛋白水平。分析miR-21、ER mRNA、PR mRNA表达水平相关性及与子宫肌瘤患者临床病理特征关系。结果miR-21、ER mRNA、PR mRNA在子宫肌瘤组患者血清中表达水平均显著高于正常子宫肌层组(P<0.05)。ER蛋白、PR蛋白在子宫肌瘤组织中阳性表达率均显著高于正常子宫肌层组织(P<0.05)。ER阴性组miR-21表达水平显著低于ER阳性组(P<0.05),PR阴性组miR-21表达水平显著低于PR阳性组(P<0.05)。子宫肌瘤患者血清miR-21及子宫肌瘤组织ER蛋白、PR蛋白表达水平与肌瘤数目、阴道不规则出血、月经周期有关(P<0.05)。子宫肌瘤患者血清miR-21表达水平与ER mRNA表达水平呈正相关(r=0.422,P<0.05),与PR mRNA表达水平呈正相关(r=0.419,P<0.05)。结论子宫肌瘤患者血清中miR-21表达水平显著升高,与肌瘤数目、阴道不规则出血、月经周期及ER、PR水平有关,可能通过影响ER、PR水平调节子宫肌瘤发生发展。Objective To investigate the expression levels of serum miR-21 in patients with uterine leiomyoma,and to analyze its correlation with estrogen receptor(ER)and progesterone receptor(PR).Methods A total of 118 patients with uterine leiomyoma who were admitted and treated in our hospital from January 2016 to January 2019 were enrolled in the study,at the same time,the other 118 patients with uterine prolapse who underwent hysterectomy were enrolled as control group.Real-time fluorescence quantitative assay(qRT-PCR)was used to detect the expression levels of miR-21,ER and PR in serum,and immunohistochemical staining was used to detect the expression levels of ER and PR in tissues.The correlation between the expression levels of miR-21,ER mRNA and PR mRNA and their correlation with clinicopathological characteristics of patients with uterine leiomyoma were analyzed.Results The expression levels of serum miR-21,ER mRNA and PR mRNA in uterine leiomyoma group were significantly higher than those in normal uterine myometrium group(P<0.05).The positive expression rates of ER protein and PR protein in uterine leiomyoma group were significantly higher than those in normal uterine myometrium group(P<0.05).The expression levels of miR-21 in ER negative group were significantly lower than those in ER positive group(P<0.05),moreover,which in PR negative group were significantly lower than those in PR positive group(P<0.05).The expression levels of miR-21 in serum and ER protein,PR protein in uterine leiomyoma tissues were closely correlated with the number of leiomyoma,irregular vaginal bleeding and menstrual cycle(P<0.05).The expression levels of serum miR-21 in patients with uterine leiomyoma were positively correlated with those of ER mRNA(P<0.05),and with those of PR mRNA(P<0.05).Conclusion The expression levels of miR-21 in serum of patients with uterine leiomyoma are significantly increased,which are related to the number of leiomyomas,irregular vaginal bleeding,menstrual cycle and those of ER and PR,which may regulate the
