摘 要:目的制备叶酸靶向载硫化铋(Bi 2 S 3)与阿霉素(DOX)相变型纳米粒(FBS-PFP-PLGA@DOX),观察其基本性能、体外多模态显像及光热联合治疗体外卵巢癌的效果。方法以双乳化法制备FBS-PFP-PLGA@DOX,检测其理化性质;观察其体外多模态显像;采用近红外激光辐照FBS-PFP-PLGA@DOX并实时记录其温度变化;以细胞计数试剂盒(CCK-8)评估光热联合治疗对卵巢癌细胞(SKOV-3)的体外杀伤作用。结果所制备FBS-PFP-PLGA@DOX平均粒径为(255.00±90.82)nm,平均电位为(-7.07±4.33)mV,DOX及Bi 2 S 3包封率分别为(26.77±0.46)%和(87.57±0.12)%。体外实验中,FBS-PFP-PLGA@DOX浓度越高,多模态显像效果越好。激光照射后FBS-PFP-PLGA@DOX迅速升温,并呈浓度依赖性。FBS-PFP-PLGA@DOX纳米粒与SKOV-3细胞孵育并辐照激光后细胞存活率为12.17%,杀伤肿瘤细胞能力高于其余各组。结论所制备FBS-PFP-PLGA@DOX具备体外多模态显像和联合治疗卵巢癌能力。Objective To prepare folate-targeted bismuth sulfide(Bi 2 S 3)and doxorubicin(DOX)phase-change poly(lactic co glycolic acid)(PLGA)nanoparticles(FBS-PFP-PLGA@DOX),and to observe its basic performance,multi-modal imaging and therapeutic effects of photothermal combined with chemotherapy for ovarian cancer in vitro.Methods FBS-PFP-PLGA@DOX were prepared by using double emulsion solvent evaporation method(W/O/W).The basic physicochemical properties and multi-modal imaging effects of FBS-PFP-PLGA@DOX were detected.The temperature variation was recorded in real time by using irradiating FBS-PFP-PLGA@DOX with near-infrared laser.The cell killing effects of photothermal therapy on ovarian cancer cell(SKOV-3)combined with chemotherapy were evaluated with Cell Counting Kit-8(CCK-8).Results The average particle size of prepared FBS-PFP-PLGA@DOX was(255.00±90.82)nm,and the average zeta potential of FBS-PFP-PLGA@DOX nanoparticles was(-7.07±4.33)mV.The entrapment efficiency of DOX and Bi 2 S 3 was(26.77±0.46)%and(87.57±0.12)%,respectively.The multi-modal imaging effects gradually enhanced with the increasing concentration of FBS-PFP-PLGA@DOX.After near-infrared laser irradiation,the nanoparticles were rapidly warmed up in a concentration-dependent manner.The cell survival rate of FBS-PFP-PLGA@DOX after laser irradiation was 12.17%,killing tumor cells better than the other groups.Conclusion The prepared FBS-PFP-PLGA@DOX had the ability of multi-modal imaging and combined treatment of ovarian cancer in vitro.
