P16基因甲基化与狼疮性肾炎的关系

摘　　要：目的研究P16基因甲基化与狼疮性肾炎的关系。方法选择2017年1月—2018年12月本院收治的26例狼疮性肾炎的患者为狼疮性肾炎组,12例系统性红斑狼疮不合并肾损伤的患者为狼疮非肾炎组,另选取年龄、性别相匹配的20名正常健康成人为健康对照组。收集三组的血液标本,提取全血DNA,甲基化特异PCR(MSP)方法检测P16基因启动子区甲基化表达情况,统计学处理实验数据。结果26例狼疮性肾炎病例组中P16基因启动子区甲基化率为61.53%(16例甲基化阳性),高于狼疮不伴肾损害甲基化率的41.67%(5例甲基化阳性)(P<0.05),两组均高于对照组的5.00%(1例甲基化阳性)。结论P16基因启动子甲基化率在狼疮性肾炎组高于狼疮不伴肾损害组,狼疮患者组明显高于健康对照组,P16基因启动子区甲基化状态可能对狼疮性肾炎的发生发展起到一定的作用。Objective To study the relationship between P16 gene methylation and lupus nephritis.Methods From January 2017 to December 2018,26 cases of patients suffered with lupus nephritis those admitted to our hospital were selected as the lupus nephritis group,and 12 cases of patients with systemic lupus erythematosus but not complicated with kidney injury were enrolled as the lupus non-renal group,and another 20 health adults were selected as health controls.To detect the promoter methylation of p16 gene,peripheral blood samples were collected for Methylation-Specific Polymerase Chain Reaction(MSP)analysis,and the results were statistically analyzed.Results According to the statistical analysis,positive methylation ratio of p16 is 61.53%(16 cases)in the patients with lupus nephritis,which was higher than those in the lupus patients without renal damage(5,41.67%)and control group(1,5.00%).Conclusions In our study,the rate of promoter methylation in lupus nephritis group was higher than that in the lupus non-renal group,and the rate in lupus non-renal group was higher than the control group.The methylation in promoter region of P16 gene may play a role in the development of lupus nephritis.
