摘 要:目的观察腺苷三磷酸(ATP)体外刺激对人外周血单个核细胞(PBMCs)人β防御素2(hBD-2)表达的影响,并通过白细胞介素1β(IL-1β)变化探讨其分子机制,为ATP提高机体抗感染能力并应用于临床提供依据。方法取健康志愿者外周血,分离PBMCs并配成细胞悬液。将PBMCs分为六组,空白组不处理,0、25、50、100、200μmol/L ATP组分别加入HBSS液和25、50、100、200μmol/L ATP 100μL,分别于培养12、24 h时收集培养上清液,采用双抗体夹心ELISA法检测上清液中的hBD-2、IL-1β。结果与空白组和0μmol/L ATP组比较,不同浓度ATP组处理的PBMCs培养上清液hBD-2、IL-1β水平不同程度的升高(P均<0.01);除空白组与0μmol/L ATP组外,其余各组培养24 h时PBMCs培养上清液hBD-2、IL-1β水平均高于同组培养12 h(P均<0.01)。经ATP作用12、24 h时,PBMCs培养上清液中hBD-2与IL-1β水平均呈正相关(r分别为0.494、0.725,P均<0.01)。结论ATP可诱导人PBMCs中hBD-2表达上调,且hBD-2与IL-1β表达变化一致;这提示ATP诱导的hBD-2表达与IL-1β密切相关,两者可能互相影响。Objective To observe the expression of human β defensin 2(hBD-2) in the human peripheral blood mononuclear cells(PBMCs) stimulated by adenosine triphosphate(ATP) in vitro,and to explore its molecular mechanism through interleukin 1β(IL-1β) changes in order to improve the body’s anti-infective ability and to provide a basis for the clinical application.Methods Peripheral blood was obtained from healthy volunteers,and PBMCs were isolated and compounded into the cell suspension.PBMCs were then divided into six groups as follows:the blank group which was not treated,and 0,25,50,50,100,and 200 μmol/L ATP groups which were added with HBSS solution and 100 μL each of25,50,50,100,and 200 μmol/L ATP;hBD-2 and IL-1β in the supernatant were collected at 12 and 24 h of incubation,respectively,and hBD-2 and IL-1β in the supernatant were detected by ELISA.Results Compared with the blank group and 0 μmol/L ATP group,the levels of hBD-2 and IL-1β in the culture supernatant of PBMCs in the ATP groups increased to various degrees(all P<0.01).Except for the blank group and 0 μmol ATP group,the levels of hBD-2 and IL-1β in the culture supernatant of PBMCs cultured for 24 h were higher than those cultured for 12 h in the same group(both P<0.01).The levels of hBD-2 and IL-1β were positively correlated in the culture supernatants of PBMCs treated with ATP for 12 or24 h(r=0.494,0.725,respectively,both P<0.01).Conclusion ATP can induce the up-regulation of hBD-2 expression in the human PBMCs,and the expression change of hBD-2 is consistent with that of IL-1β,suggesting that ATP-induced hBD-2 expression is closely related to IL-1β,and they may interact with each other.
