摘 要:目的:分析HSP70调控急性脓毒症患者内皮细胞损伤的机制。方法:选择2017年2月~2018年12月本院收治的3例急性脓毒症患者作为观察组,3例行手术治疗的骨折患者作为对照组;提取受试者内皮细胞,分为空白对照组及10、50、100 mg/L组,采用MTT检测各组细胞活力,采用荧光显微镜观察细胞核形态,并通过透射电镜观察内皮细胞自噬体,后检测Bcl-2、Beclin-1及β-actin蛋白表达量。结果:HSP70的干预可有效改善急性脓毒症患者内皮细胞活力,其中观察组和对照组中100 mg/L亚组细胞活力最高,空白对照亚组细胞活力最低,差异具有统计学意义(P<0.05);相较于对照组,急性脓毒症患者内皮细胞细胞核缺损严重,采用HSP70的干预可有效改善细胞核形态、自噬体形态和结构形态,其中100 mg/L亚组的细胞核形态、自噬体形态和结构最佳;HSP70干预可有效改善急性脓毒症患者内皮细胞内Bcl-2及Beclin-1水平,其中观察组和对照组中的100 mg/L亚组中Bcl-2及Beclin-1水平最高,空白对照亚组中最低,且差异有统计学意义(P<0.05)。结论:HSP70可有效调控急性脓毒症患者内皮细胞中Bcl-2水平,有效抑制细胞凋亡并缓解细胞皮肤损伤。Objective:To analyze the mechanism of HSP70 in regulating endothelial cell injury ofpatients with acute sepsis.Methods:Three patients with acute sepsis admitted to our hospital from February 2017 to December 2018 were selected as the observation group,and three patients with surgical fractures were used as the control group.Endothelial cells were extracted and divided into a blank control group,10 mg/L,50 mg/L and 100 mg/L groups.MTT was used to detect the cell viability,fluorescence microscopy was used to observe nuclear morphology,and endothelial cell autophagosomes were observed by transmission electron microscopy.Protein expression of Bcl-2,Beclinandβ-actin were detected.Results:The intervention of HSP70 can effectively improve the viability of endothelial cells in patients with acute sepsis.The 100 mg/L group and the control group had highercell viability,whilethe blank control group had the lowest cell viability,and the difference was statistically significant(P<0.05).Compared with the control group,patients with acute sepsis hadsevere endothelial cell nuclear defects.The intervention of HSP70 couldeffectively improve the nuclear morphology,autophagosome morphology and structural morphology,of which the 100 mg/L subgroup had the best effect.HSP70 intervention couldeffectively improve the levels of Bcl-2 and Beclin-1 in endothelial cells of patients with acute sepsis.Bcl-2 and Beclin-1 were higher in the 100 mg/L group and control group while the blank control group is the lowest,and the difference wasstatistically significant(P<0.05).Conclusion:sHSP70 can effectively regulate the level of Bcl-2 in endothelial cells of patients with acute sepsis,effectively inhibit apoptosis and relieve cell skin damage.
