稳心颗粒对心肌梗死大鼠模型心肌脂肪化及E6AP、C/EBPα表达影响

摘　　要：目的:通过观察稳心颗粒对大鼠心肌细胞诱导分化成前脂肪细胞E相关蛋白(E6 assiociated Protein,E6AP)、转录因子CCAAT/增强子结合蛋白(C/EBPα)表达的影响,探讨稳心颗粒对心肌脂肪化作用机制。方法:将大鼠心肌细胞诱导分化成前脂肪细胞,分成空白组、E6AP腺病毒组、E6AP封闭组、E6AP过表达组、胺碘酮组、稳心颗粒组,相应组别给予腺病毒感染及含药血清处理培养,采用RT-PCR和免疫组化检测各组E6AP、C/EBPα表达,同时通过油红染色对比各组前脂肪细胞脂肪化情况。结果:E6AP过表达组E6AP表达明显高于各组而C/EBPα表达显著低于各组,E6AP与C/EBPα表达具有负性调节关系,稳心颗粒组E6AP表达显著高于胺碘酮组但C/EBPα表达2组无明显差异,E6AP封闭组油红染色程度最深而E6AP过表达组染色明显减少,胺碘酮组和稳心颗粒组染色程度相对于空白组有较为明显的减少。结论:稳心颗粒可能通过上调E6AP和抑制C/EBPα表达抑制心肌脂肪化。Objective To observe the effect of Wenxin Granule on the expression of E-related protein(E6 assiociated Protein,E6AP)and enhancer-binding protein(C/EBPα)of preadipocytes induced by rat cardiomyocytes,and to explore the mechanism of Wenxin Granule on myocardial adipogenesis.Methods Rat cardiomyocytes were induced to differentiate into preadipocytes and divided into blank group,E6AP adenovirus group,E6AP blocking group,E6AP overexpression group,amiodarone group,and Wenxin granule group.And the corresponding groups were given adenovirus infection and drug-containing Serum-treated cultures were used to detect the expression of E6AP and C/EBPαin each group by RT-PCR and immunohistochemistry.At the same time,the fat metaplasia of preadipocytes in each group was compared by oil red staining.Results The expression of E6AP in the E6AP over-expression group was significantly higher than that in each group and the expression of C/EBPαwas significantly lower than that in each group.The expression of E6AP and C/EBPαhad a negative regulatory relationship.The expression of E6AP in Wenxin Granule Group was significantly higher than the amiodarone group,but there was no significant difference in C/EBPαexpression between the two groups.The degree of oil red staining was the deepest in the E6AP blocking group,while the staining in the E6AP overexpression group was significantly reduced.And compared with the blank group,the degree of staining in the amiodarone group and Wenxin Granule group has a more obvious reduction.Conclusion Wenxin Granule may inhibit myocardial adipogenesis by upregulating E6AP and inhibiting the expression of C/EBPα.
