摘  要:本研究利用高能和低能饲粮饲喂育成期蛋鸡,运用small RNA测序方法对肝脏中miRNA进行检测,对其表达量进行分析,对差异表达miRNA进行靶基因预测,并对差异表达的预测靶基因进行基因本体论(GO)功能和京都基因与基因组百科全书(KEGG)信号通路富集分析。结果表明:与低能组相比,高能组有2个miRNA表达显著上调(P<0.05),6个miRNA表达显著下调(P<0.05);在差异表达基因中共预测出差异表达miRNA的41个靶基因,其中miR-15a的预测靶基因最多,为26个;差异表达靶基因GO和KEGG分析发现,免疫和分子功能调控是富集程度最高的过程,而显著富集的信号通路与免疫和脂质代谢相关。由此可见,能量摄入量通过影响miRNA的表达进而上调或下调肝脏中免疫和脂质代谢相关靶基因的表达,最终调控蛋鸡的免疫和脂质代谢,并影响生产性能。In current study,miRNAs in liver were detected by small RNA sequencing of laying hens fed with high-energy or low-energy during the growing period,and their expression levels were analyzed.Target genes of differentially expressed miRNAs were predicted,and differentially expressed target genes were used to undergo gene ontology(GO)and kyoto encyclopedia of genes and genomes(KEGG)enrichment analysis.The results showed that compared with low energy group,the expression levels of 2 miRNAs in high energy group were up-regulated and 6 miRNAs were down-regulated(P<0.05).Forty target genes of differentially expressed miRNAs were predicted in differentially expressed genes,and the number of predicted target genes of miR-15a was the biggest for 26.Analysis of GO and KEGG of differentially expressed target genes showed that immune and regulation of molecular function were the most enriched processes,and the significantly enriched signaling pathways were related with immune and lipid metabolism.It is indicates that energy intake regulates immune and lipid metabolism of laying hens by influencing expression of miRNAs and then upregulates or downregulates expression of immune and lipid metabolism related target genes in liver,and finally affects production performance.
