摘  要:目的通过动物实验和网络预测的形式来揭示龙生蛭胶囊中“黄芪-当归-刺五加”药组治疗脑卒中后PSCI(PSCI)的作用机制,为龙生蛭胶囊的开发研究及临床应用提供参考。方法采用Morris水迷宫实验评价“黄芪-当归-刺五加”药组对PSCI大鼠行为学的影响,同时对血清生化学相关指标进行测定和比较;采用整合药理学平台(TCMIP V2.0)对“黄芪-当归-刺五加”药组治疗PSCI的作用机制进行挖掘与预测。结果“黄芪-当归-刺五加”药组对PSCI大鼠的空间记忆能力和学习能力具有良好的改善作用;同时药组还能显著降低模型脑卒中大鼠血清中丙二醛(MDA)的含量,提高超氧化物歧化酶(SOD)的活性。共预测得到27个活性成分,11个药物与疾病的共有靶标。结论证实了“黄芪-当归-刺五加”对PSCI大鼠的行为学及血清生化指标具有一定的影响及改善作用,同时也初步预测了“黄芪-当归-刺五加”药组治疗PSCI的药效物质基础及作用机制,可为龙生蛭胶囊的进一步开发利用提供参考。Objective To analyze the mechanism of Astragalus-Angelica sinensis-Schisandra chinensis drug group in Longshengzhi capsules for post-stroke cognitive impairment(PSCI)based on animal experiments and network pharmacology,and to provide reference for the development and clinical application of Longshengzhi capsules.Methods Morris water maze test was used to evaluate the effect of Astragalus-Angelica sinensis-Schisandra chinensis drug group on the behavior of PSCI in rats,and then the serum biochemical indicators were measured and compared.At the same time,based on the integrative pharmacology of traditional Chinese medicine(TCMIP V2.0),the mechanism of Astragalus-Angelica sinensis-Schisandra chinensis drug group for PSCI was explored.Results The experimental showed that the Astragalus-Angelica sinensis-Schisandra chinensis drug group improved the spatial memory and learning ability of PSCI in rats.The drug group also significantly reduced the content of MDA and increased the activity of SOD in the serum.Totally 27 compounds and 11 shared targets for drug and disease.Conclusion Astragalus-Angelica sinensis-Schisandra chinensis drug group can improve the behavioral and serum biochemical indexes for PSCI rats by experiment.Meanwhile,this study predicts the bioactive constituents and relative molecular mechanism of the drug group for PSCI,and lays a solid foundation for its further exploitation and utilization.
