摘  要:目的探讨理气通便合剂对便秘型肠易激综合征(IBS-C)大鼠的可能作用机制。方法将SD大鼠24只按随机数字表法分为空白组、IBS-C模型组、理气通便合剂组、莫沙必利组,每组6只。采用冰水灌胃法制备IBS-C模型。造模7天后,予药物干预,理气通便合剂组予理气通便合剂(18.75g·kg^-1·d^-1)灌胃,莫沙必利组予莫沙必利水溶液(1.35mg·kg^-1·d^-1)灌胃,空白组及IBS-C模型组给予等量蒸馏水灌胃。21天后摘取各组大鼠下丘脑及结肠组织,酶联免疫吸附试验(ELISA)检测5-羟色胺(5-HT)、血管活性肠肽(VIP)表达水平。结果与空白组比较,IBS-C模型组大鼠结肠5-HT[(35.94±2.06)ng/g比(25.88±3.10)ng/g]、下丘脑组织5-HT[(71.12±5.88)ng/g比(50.85±3.14)ng/g]、结肠组织VIP[(656.37±60.34)pg/g比(577.86±53.55)pg/g]、下丘脑组织VIP[(386.57±38.00)pg/g比(276.17±43.73)pg/g]均明显升高(P<0.05),药物干预后,理气通便合剂组及莫沙必利组大鼠结肠及下丘脑组织5-HT、VIP表达水平较IBC-C模型组有所下降,理气通便合剂组较模型组差异有统计学意义(P<0.05),莫沙必利组无明显统计学差异(P>0.05)。结论理气通便合剂治疗IBS-C作用可能是通过抑制5-HT、VIP,调节肠道运动及分泌功能实现的。Objective To investigate the effects of Chinese medicine Li Qi Tong Bian decoction in controlling rat model of constipation-predominant irritable bowel syndrome(IBS-C)and the underlying molecular mechanism.Methods Twenty-four Sprague Dawley rats were randomly divided into control,IBS-C model,treatment(Chinese medicine),and mosapride(Western medicine)groups.IBS-C model was established using ice-water gavage.Seven days after modeling,mice in the treatment group were administered with Li Qi Tong Bian decoction at 18.75g/kg daily and in the Western medicine group were administered with mosapride aqueous solution at a daily dose of 1.35mg/kg.The control and IBS-C rats were given an equal amount of distilled water for 21 days.After that,the hypothalamus and colon tissues of each rat were taken for analysis of 5-hydroxytryptamine(5-HT)and vasoactive intestinal peptide(VIP)expression using ELISA.Results Compared with that of control rats,levels of colonic and hypothalamus 5-HT protein in the model rats were significantly increased(35.94±2.06ng/g vs.25.88±3.10ng/g and 71.12±5.88ng/g vs.50.85±3.14ng/g,respectively;P<0.05)and VIP protein also showed a similar trend(656.37±60.34pg/g vs.577.86±53.55pg/g and 386.57±38.00 pg/g vs.276.17±43.73pg/g,respectively;P<0.05).After drug intervention,levels of 5-HT and VIP proteins were dramatically reduced in rat colon and hypothalamus in both traditional Chinese medicine and Western medicine groups with statistical difference between Chinese medicine and the model rats,while without statistical difference between Western medicine and the model rats.Conclusion The therapeutic effect of Li Qi Tong Bian decoction can reduce levels of 5-HT and VIP,indicating that this decoction in regulating intestinal movement and secretory function may be through 5-HT and VIP suppression.
