摘  要:目的:采用HPLC和NIRS分析技术,分别建立三七花、人参花、西洋参花指纹图谱,并比较分析研究三七花、人参花、西洋参花指纹图谱之间的差异,提供可靠的品种鉴别依据。方法:考察建立样品前处理方法,建立HPLC色谱条件,分别收集三七花、人参花、西洋参花HPLC图谱,通过比较对照品色谱峰保留时间,指认色谱峰,建立指纹图谱,分析比较各品种指纹图谱差异特征;应用FOSS NIRS,设置参数,采集850~2 499 nm色谱图,采用WinISIⅡ近红外分析软件建立各品种近红外原始指纹图谱、一阶导数处理指纹图谱、二阶导数处理指纹图谱,分析比较各品种指纹图谱差异特征。结果:比较分析各品种的HPLC、NIRS指纹图谱特征,发现3种样品的化学成分存在西洋参花成分涵盖了人参花成分,人参花成分涵盖了三七花成分的关系。可通过HPLC指纹图谱中色谱峰S29、S31、R20、R21、R22、R32鉴别三七花与人参花,通过色谱峰X23、X24、X25、X35、X36、X40鉴别三七花与西洋参花,通过R27、X21、X34、X36鉴别人参花与西洋参花;二阶导数处理图谱与原始图谱和一阶导数处理图谱相比,三七花、人参花、西洋参花图谱峰数增加,峰形更加明显,能更好地显示出3个品种之间的差异特征。可通过二阶导数处理指纹图谱1 690~1 726nm、2 260~2 308 nm色谱峰形鉴别三七花与人参花、西洋参花,二阶导数处理指纹图谱2 328~2 346 nm的峰形鉴别西洋参花与人参花。结论:三七花、人参花、西洋参花HPLC指纹图谱、近红外指纹图谱存在差异特征,HPLC指纹图谱相比NIRS指纹图谱能更好的体现出3个品种之间的差异特征。本文建立的指纹图谱鉴别方法为三七花、人参花、西洋参花进一步的鉴别研究与研究开发提供了可靠参考依据。Objective:To establish the fingerprint of P. notoginseng flower,P. ginseng flower and P. quinquefolium flower by HPLC and NIRS analysis technoloyies,and to compare and analyze the differences among them. Methods:By comparing the retention time of reference substances peaks,identified peaks,established fingerprints,the different characteristics of fingerprint of each variety were analyzed and compared. The WinISI Ⅱ NIRS analysis software was used to establish the original NIRS fingerprint map of each variety,the first-order and the second-order derivative processing fingerprint map,to analyze and compare the different characteristics of the fingerprint map of each variety. Results:The HPLC and NIRS fingerprint characteristics of each variety were compared and analyzed. And it was found that the chemical compositions of American ginseng flower extended beyond ginseng flower,and the chemical compositions of ginseng flower extended beyond notoginseng flower. Flowers of notoginseng and ginseng could be identified by chromatographic peaks S29,S31,R20,R21,R22 and R32 in HPLC fingerprint. Flowers of notoginseng and quinquefolium could be identified by chromatographic peaks X23,X24,X25,X35,X36 and X40. Flowers of ginseng and quinquefolium could be identified by chromatographic peaks R27,X21,X34 and X36. Compared with the original and the first derivative,the peak number and peak shape of the second derivative processing graph could better show the differences among the three varieties. Notoginseng flower and ginseng flower could be identified by the fingerprint analysis with the second derivative at 1 690-1 726 nm and 2 260-2 308 nm,and quinquefolium flower and ginseng flower could be identified by the fingerprint analysis with the second derivative at 2 328-2 346 nm. Conclusion:There are differences among HPLC and NIRS fingerprints of the three varieties,and compared with NIRS fingerprint,HPLC fingerprint can better reflect the differences among them.
