七氟烷对肺腺癌细胞增殖和迁移能力的影响

摘　　要：目的研究七氟烷对肺腺癌细胞增殖和迁移能力的影响及相关机制。方法体外培养对数生长期肺腺癌细胞系H1299,将H1299细胞传代并分为对照组及实验组,对照组用含5%CO2常规气体培养,实验组用2.5%七氟烷及5%CO2气体培养,处理细胞6 h。以TRIzol法收集2组细胞总RNA,以逆转录实时荧光定量聚合酶链反应检测2组细胞缺氧诱导因子-1α(Hif-1α)mRNA及微小RNA-210(miR-210)表达情况。收集2组细胞总蛋白,以蛋白质印迹实验检测2组细胞HIF-1α蛋白表达情况;以平板克隆形成实验及MTT法检测2组细胞的增殖能力,以Transwell实验检测2组细胞的侵袭迁移能力。结果实验组和对照组Hif-1αmRNA的表达水平分别为0.19±0.04,0.37±0.07,miR-210表达水平分别为0.06±0.02,0.14±0.03,HIF-1α蛋白表达水平分别为0.16±0.05,0.33±0.08,平板克隆形成数(个)分别为31.92±7.34,64.54±8.02,侵袭细胞数分别为(78.82±10.61),(164.08±17.34)个,迁移细胞数分别为(141.52±21.33),(224.75±26.94)个,差异均有统计学意义(均P<0.05)。结论七氟烷可通过下调Hif-1α/miR-210的表达,抑制肺腺癌细胞的增殖及迁移能力。Objective To investigate the effect of sevoflurane on the proliferation and metastasis ability of lung adenocarcinoma cells and its related mechanism.Methods The H1299 logarithmic lung adenocarcinoma cell line H1299 was cultured in vitro.The H1299 cells were passaged and divided into control group and experimental group.Control group was cultured with conventional gas containing 5%CO2.Experimental group was clutured with 2.5%sevflurane and 5%CO2,cells were treated for 6 h.TRIzol method was used to collect the total RNA of two groups of cells,and detect the hypoxia inducible factor-1α(Hif-1α)mRNA and microRNA-210(miR-210)by quantitative reverse transcription PCR.The total protein in two groups of cells were collected and the expression of HIF-1αprotein was detected by Western Blot.The plate colony formation test and MTT test were used to detect the proliferation in two groups,and the Transwell test was used to detect the invasion and migration in two groups.Results The expression level of Hif-1αmRNA in experimental group and control group were 0.19±0.04,0.37±0.07,the expression of miR-210 were 0.06±0.02,0.14±0.03,the expression of HIF-1αprotein were 0.16±0.05,0.33±0.08,the number of colony formation were 31.92±7.34,64.54±8.02,the number of invasive cells were 78.82±10.61,164.08±17.34,the number of migrating cells were 141.52±21.33,224.75±26.94,all with significant difference(all P<0.05).Conclusion Sevoflurane can inhibit the proliferation and metastasis of lung adenocarcinoma cells by down-regulating the expression of Hif-1α/miR-210.
