摘 要:目的建立测定万应跌打酒中士的宁含量的高效液相色谱法。方法色谱柱为Agilent TC-C18柱(250 mm×4.6 mm,5μm),流动相为0.01 moL/L庚烷磺酸钠与0.02 mo L/L磷酸二氢钾等量混合溶液(用10%磷酸调pH至2.8)-乙腈(79∶21,V/V),检测波长为260 nm;流速为1.0 m L/min,柱温为25℃,进样量为10μL。结果士的宁质量浓度在13.92~114.02μg/m L(r=0.9997,n=5)范围内与峰面积线性关系良好;精密度、稳定性、重复性试验的RSD均小于2%;平均加样回收率为91.48%,RSD为1.88%(n=6);样品中士的宁的最高限度应为每1 m L不得超过100μg。结论该方法简便,专属性强,重复性好,可用于测定万应跌打酒中士的宁的含量。Objective To establish a high-performance liquid chromatography(HPLC)method for the content determination of strychnine in Wanying Dieda Wine.Methods The Agilent TC-C18 column(250 mm×4.6 mm,5μm)was adopted,the mobile phase was 0.01 mol/L sodium heptane sulfonate mixed with 0.02 mol/L potassium dihydrogen phosphate in equal volume(adjusted pH to 2.8 with 10%phosphoric acid)-acetonitrile(79∶21,V/V),the detection wavelength was 260 nm,the flow rate was 1.0 m L/min,the column temperature was 25℃,and the injection volume was 10μL.Results Strychnine had good linearity in the range of 13.92-114.02μg/m L(r=0.9997,n=5).The RSDs of precision,stability and repeatability tests were less than 2%,the average recovery rate was 91.48%with RSD of 1.88%(n=6).The maximum limit of strychnine in Wanying Dieda Wine should not exceed 100μg/m L.Conclusion The method is simple,specific and reproducible,which can be used for the content determination of strychnine in Wanying Dieda Wine.
