摘  要:试验旨在研究通过16S rDNA高通量测序分析技术测定饲粮中添加γ-聚谷氨酸(γ-PGA)对小尾寒羊瘤胃发酵参数及菌群结构的影响。采用单因素完全随机试验设计,选取检疫合格、体重(37.1±0.5)kg的3月龄小尾寒羊公羊24只,随机分为对照组和试验组。对照组饲喂基础饲粮,试验组在基础饲粮中添加20 g/(只·d)的γ-PGA,预试期14 d,正试期7 d,正试期第7天晨饲2 h后通过瘤胃液采样器采集瘤胃液测定瘤胃发酵参数,并利用16S rDNA高通量测序分析技术测定瘤胃液中菌群结构。结果表明:试验组小尾寒羊瘤胃液氨态氮(P<0.01)、乙酸浓度(P<0.05)高于对照组,但两组试验羊的瘤胃液pH、丙酸、丁酸、乙酸/丙酸及微生物蛋白间无显著差异;饲粮中添加γ-PGA可提高小尾寒羊瘤胃菌群多样性(P<0.01);在门水平上,试验组小尾寒羊瘤胃液中软壁菌门、螺旋菌门、未注释细菌门及蓝藻门的相对丰度低于对照组(P<0.01),放线菌门的相对丰度高于对照组(P<0.01);试验组拟杆菌门和互养菌门的相对丰度均高于对照组(P>0.05);在属水平上,饲粮添加γ-PGA后小尾寒羊瘤胃液中普雷沃式菌属的相对丰度提高了8.2%,并且可提高小尾寒羊瘤胃内解琥珀酸菌属的相对丰度(P<0.01)。由此可见,饲粮中添加γ-PGA能够极显著提高小尾寒羊瘤胃乙酸、氨态氮及菌群多样性,对其纤维素降解菌的生长影响显著,可提高拟杆菌门和互养菌门的相对丰度,还能够提高普雷沃式菌属的相对丰度。The present study was conducted to examine the effect of dietary supplementation of poly-γ-glutamate(γ-PGA)on rumen fermentation parameters and microflora structure of Small Tail Han Sheep based on 16S rDNA high-throughput sequencing technology.Twenty-four Small Tail Han Sheep of 3 months old with similar quarantine and body weight were randomly divided into two groups.The sheep in control group were fed with basal diet and that in treat group were fed with the basal diet with 20 g/(d•sheep)γ-PGA.The experiment adopted one–factor completely random design with 21 d(14d for pre-experiment and 7 d for test).The rumen fluid samples were collected at 2 h after feeding on d 7 for test.The 16S rDNA high-throughput sequencing technique was used to determine the bacterial community structure in rumen fluid.The results showed that ammonia nitrogen in treat group was extremely higher than that in control group(P<0.01);and the concentration of acetic acid in treat group was significantly higher than that in control group;However,there was no significant difference in pH,propionic acid,butyric acid,acetic acid/propionic acid and microbial protein between two groups(P>0.05).Compared with control group,addingγ-PGA significantly increased bacterial diversity in rumen(P<0.01).At the phylum level,compared with control group,the relative abundance of Tenericutes,Spirochaetae,Cyanobacteria and the Unclassified Bacteria Phylum in treat group decreased significantly(P<0.01),and that of Bacteroidetes and Synergistetes increased by 6.3%and 3.6%,respectively.At the genus level,compared with control group,the relative abundance of Succiniclasticum in treat group increased significantly(P<0.01),and that of Prevotella_1 increased by 8.2%.Overall,in this experiment,addingγ-PGA increased the acetic acid,ammonia nitrogen and the diversity of rumen bacteria of Small Tail Han Sheep,which obviously affect the growth of cellulolytic bacteria flora and increase the relative abundance of Bacteroidetes,Synergistetes,and Prevotella_1 in rumen
