摘  要:目的:建立茯苓饮片水煎液中多糖的含量测定方法,并对不同规格、不同煎煮时间下茯苓水煎液中多糖含量进行测定和比较,为中医临床用药提供参考。方法:采用苯酚-硫酸法联合3,5-二硝基水杨酸法测定茯苓块和茯苓片在不同煎煮时间下水煎液中茯苓多糖的含量。结果:苯酚-硫酸法在490nm波长下吸光度与总糖含量呈良好线性关系,线性范围为0.02~0.10μg/mL,r=0.9997,加样回收率为102.70%;3,5-二硝基水杨酸法在540nm波长下吸光度与单糖含量呈良好线性关系,线性范围为0.20~0.60μg/mL,r=0.9999,加样回收率为101.06%。随着煎煮时间的增加,茯苓水煎液中茯苓多糖含量也持续增加,45min时茯苓多糖成分基本煎出完全。结论:建立的多糖测定方法简单、准确、适用性好,可用于茯苓饮片水煎液中茯苓多糖含量测定。结合实验结果及临床实际情况,最佳煎煮时间为45min。Objective To establish a method for quantitative determination of polysaccharide in the decoction of-Poriacocospieces,and to determine and compare the content of polysaccharide in different specification and different decoction times.Methods Phenol-sulfuric acid method and 3,5-dinitrosalicylic acid method were used to determine the content of polysaccharides in different specification and different decoction times.ResultsThe phenolic-sulfuric acid method has a good linear relationship with the total sugar content at 490nm wavelength,with a linear range of 0.02~0.10μg/mL,r=0.9997,theaverage recovery was 102.70%;The 3,5-dinitrosalicylic acid method has a good linear relationship with the monosaccharide content at 540nm wavelength,with a linear range of 0.20~0.60μg/mL,r=0.9999,theaverage recovery was101.06%.With the increase of decoction time,the content of polysaccharides in the decoctionliquid of the pachymaran also continued to increase,45min when the polysaccharides are basically decoction completely.ConclusionThe method of polysaccharides is simple,accurate and well applicable,and can be used to determine the polysaccharide content in the water decoction solution of the saccharine.Combined with the experimental results and clinical actual situation,the optimal decoction time is 45min.
