摘  要:目的探究丙泊酚对七氟烷引起的大鼠海马神经细胞损伤及认知障碍的相关机制。方法60只SD大鼠随机分为对照组、七氟烷组、联合组,每组20只。七氟烷组大鼠每天吸入1.5%七氟烷1 h;联合组在七氟烷组基础上每天腹腔注射丙泊酚注射液150 mg/kg;对照组腹腔注射等量生理盐水。比较3组大鼠神经功能及海马细胞凋亡率、凋亡相关蛋白Bax、Bcl-2、Caspase-3表达及血清炎性因子[白介素-6(IL-6)、IL-1β、肿瘤坏死因子-α(TNF-α)]含量水平。结果与对照组相比,七氟烷组Y-迷宫逃离时间、错误反应次数、Longa评分、Bax和Caspase-3蛋白相对表达水平、海马区细胞凋亡率、血清IL-6、IL-1β和TNF-α含量显著升高,Bcl-2蛋白相对表达水平显著降低(P<0.05);与七氟烷组相比,联合组Y-迷宫逃离时间、错误反应次数、Longa评分、Bax和Caspase-3蛋白相对表达水平、海马区细胞凋亡率、血清IL-6、IL-1β和TNF-α含量水平显著降低,Bcl-2蛋白相对表达水平显著升高(P<0.05)。结论丙泊酚可以缓解七氟烷引起的大鼠海马神经细胞损伤,其作用机制与抑制海马细胞凋亡和炎症反应相关。Objective To explore the effects of propofol on hippocampal neuronal apoptosis and cognitive impairment in rats induced by sevoflurane(Sevo)and its related mechanism.Methods Sixty SD rats were randomly divided into control group,Sevo group and Sevo combined with propofol group,with 20 rats in each group.The rats in the Sevo group inhaled 1.5%Sevo for 1 h daily;the Sevo combined with propofol group was intraperitoneally injected with propofol 150 mg/kg daily on the basis of Sevo group;the control group was intraperitoneally injected with the same amount of normal saline.Neurological function,apoptosis rate of hippocampal cells,expression of apoptosis related proteins Bax,Bcl-2 and Caspase-3,and serum levels of inflammatory factors[interleukin(IL-6),IL-1β,tumor necrotic factor(TNF)-α]were compared among the three groups.Results Compared with the control group,escape latency in Y-maze test,number of false reactions,Longa score,relative expression levels of Bax and Caspase-3 protein,hippocampal cell apoptosis rate,content of serum IL-6,IL-1β,and TNF-αwere significantly increased in the Sevo group,and the relative expression level of Bcl-2 protein was significantly decreased(P<0.05).Compared with the Sevo group,escape latency in Y-maze test,number of false reactions,Longa score,relative expression levels of Bax and Caspase-3 protein,hippocampal cell apoptosis rate,content of serum IL-6,IL-1β,and TNF-αwere significantly decreased in the Sevo group,and the relative expression level of Bcl-2 protein was significantly increased(P<0.05).Conclusion Propofol can alleviate hippocampal neuronal injury induced by Sevo in rats,and its mechanism of action is related to inhibiting hippocampal cell apoptosis and inflammatory response.
