摘  要:目的:探讨6种asm基因表达水平与安丝菌素P-3(AP-3)产量的相关性。方法:发酵培养珍贵束丝放线菌ATCC31565,高效液相色谱法检测发酵液中AP-3的产量,实时荧光定量PCR检测asm基因表达量并与AP-3产量进行相关性比对。结果:asm2和asm36基因表达量随培养时间增加而升高,与AP-3产量呈显著的正相关性(P<0.001);asm20和asm39基因表达量在生产后期显著升高,与AP-3产量具有正相关性(P<0.05);asm28和asm30基因表达量与AP-3产量没有显著相关性。结论:asm2和asm36基因表达量与AP-3产量之间存在正相关性,提示该基因对AP-3合成的正向调控功能,为今后提高AP-3产量的合成途径改造提供了候选基因,为后续AP-3产量的提高奠定了基础。Objective:To investigate the correlation between the expression levels of six asm genes and the yield of ansamitocin P-3(AP-3).Methods:The production of AP-3 in fermentation broth of Actinosynnema pretiosum ATCC31565 was determined by using high performance liquid chromatography(HPLC),and the expression of asm genes was determined by real-time fluorescent quantitative PCR,and the correlation between AP-3 production and the expression of asm genes was compared.Results:The expression levels of asm2 and asm36 increased with the increase of culture time,and had a significant positive correlation with AP-3 yield(P<0.001);the expression lev⁃els of asm20 and asm39 increased significantly in the late stage of fermentation,and had a positive correlation with the yield of AP-3(P<0.05).However,the expression of asm28 and asm30 had no significant correlation with the yield of AP-3.Conclusion:There is a positive correlation between the expression levels of asm2 and asm36 genes and the yield of AP-3,which indicates that asm2 and asm36 genes have a positive regulatory function on AP-3 synthesis,which provides candidate genes for improving AP-3 production in the future,and lays a founda⁃tion for the improvement of AP-3 yield in the future.
