摘  要:研究染料茜素绿-蛋白质-Cu(Ⅱ)三元络合物的共振瑞利散射光谱,建立共振瑞利散射光谱测定蛋白质的新方法。在pH为4.35的B-R缓冲溶液下,茜素绿与Cu(Ⅱ)之间相互作用,形成配位比4∶1的配合物,再与蛋白质作用形成三元络合物,使被测物的共振瑞利散射(RRS)强度显著增强,在一定范围内相对散射强度ΔI与蛋白质含量成正比。实验确定了反应的最佳条件,并对牛奶、豆浆两种样品进行测定,结果满意。It was studied that a new method for the determination of proteins by the dye-metal probe composed of dye alizarin green and metal ion Cu(Ⅱ).in the B-R buffer solution with pH 4.35,alizarin green interacts with Cu(Ⅱ)to form a complex with a coordination ratio of 4∶1,which then interacts with proteins to form a ternary complex.This result in a significant increases in the intensity of the resonance Rayleigh scattering(RRS)of the analyte,and the relative scattering intensity ΔI is proportional to the protein content within a certain range.In the experiment,the best reaction conditions and two standard curves were determined.Milk and soybean milk samples were also determined to verify the accuracy of the method.It has also been proved that this new method for determining proteins has high sensitivity,wide application range and strong stability.Therefore,this method has a good development prospect in protein determination.
