摘 要:【目的】插穗生根是一个复杂的生理过程,而生理生化指标在插穗生根过程中发挥着重要作用。树头菜应用广泛,研究ABT-1处理对树头菜扦插生根及生理生化特性的影响,探究树头菜扦插生根机制,为其建立高效的扦插繁殖体系提供理论依据。【方法】以树头菜1年生枝条为试验材料,选用ABT-1(0,50,100,150 mg/L)溶液分别浸泡插穗3 h,探究其对扦插生根以及生理生化指标的影响。【结果】树头菜扦插后30 d左右有愈伤组织产生,45 d左右开始有不定根形成,90 d生长稳定,生根类型既有愈伤组织生根型,又有皮部生根型,且多为愈伤组织生根型,ABT-1处理各水平在8个生根指标中基本均以150 mg/L浸泡3 h效果最佳,生根率、平均根数、平均根长、最长根长、平均根粗、最大根系直径、生根指数、根系效果指数分别为82.2%、12.11条/株、17.96 cm、40.55 cm、3.59 mm、7.97 mm、13.57和1.12,且均显著(P<0.05)高于清水处理(CK)。ABT-1处理组和CK可溶性糖含量呈现先下降后上升趋势,且整个过程中ABT-1处理组可溶性糖含量始终高于CK,而处理组和CK可溶性蛋白含量总体上呈现先上升后下降再上升的趋势,ABT-1处理组含量始终高于CK;ABT-1处理组和清水处理(CK)POD活性总体上变化趋势相同,在根原基诱导期和和愈伤组织形成期,以及不定根伸长期出现1个峰值,且整个过程中处理组POD活性始终高于CK;ABT-1处理组前期SOD、CAT活性变化趋势与CK相反,后期处理组和对照组SOD活性总体上变化趋势相同,此外,处理组SOD活性始终高于对照组;ABT-1处理组和对照组(CK)MDA含量总体上呈现先小幅上升后下降最后趋于平稳趋势,且整个过程中处理组MDA含量始终小于对照组(CK)。【结论】ABT-1处理有利于插穗生根和改善根系质量,提高插穗内营养物质含量、相关酶活性,以及通过调控插穗内抗氧化酶保护系统活性变化来抑制插穗体内毒害�[Objective]Rooting of cuttings is a complex physiological process,and physiological and bio⁃chemical indexes play an important role in the process of cutting rooting.Crateva unilocalaris is widely used.This study was to explore the effect of ABT-1 treatment on rooting and physiological and biochemical charac⁃teristics of Crateva unilocalaris cuttings,and explore the rooting mechanism,so as to provide a theoretical basis for establishing an efficient cutting propagation system.[Method]The 1a branches of Crateva unilocalaris were used as the experimental materials,they were soaked in ABT-1 solution(0,50,100,150 mg/L)for 3 hours to explore its effect on cutting roots and physiological and biochemical indexes.[Result]Callus appeared about 30 days after cutting,adventitious roots began to form about 45 days,and grew steadily after 90 days.Both cal⁃lus and skin rooting were found,and most of them were callus rooting.Among the 8 rooting indexes of the ABT-1 treatment,the effect of soaking at a concentration of 150 mg/L for 3 h was the best.The rooting rate,average root number,average root length,longest root length,average root diameter,maximum root diameter,rooting in⁃dex,and root effect index were 82.2%,12.11 per plant,17.96 cm,40.55 cm,3.59 mm,7.97 mm,13.57,1.12,respectively.And all of them were significantly higher than those of CK treated with clean water.On the whole,the soluble sugar content of the ABT-1 treatment group and CK decreased at first and then increased,and dur⁃ing the whole process,the soluble sugar content of the ABT-1 treatment group was always higher than that of CK.The soluble protein content of the treatment group and CK increased at first,then decreased and then in⁃creased,and the content of the ABT-1 treatment group was always higher than that of CK.There was a peak of the changing trend of POD of CK,in the root primordium induction stage,the callus formation stage and adven⁃titious root formation and elongation stage,and the POD activity ot the ABT-1 treatment group was always hi
