摘  要:【目的】近年来,研究发现FSHR基因在非性腺组织中也有表达,能够介导FSH信号参与调控机体内多项生理活动。FSH-FSHR信号与脂肪沉积、体质量存在一定关联,但有关FSHR基因与兔体质量性状的关联还未见报道。【方法】本研究以天府黑兔为研究对象,采用PCR产物直接测序的方法对其FSHR基因第10外显子进行SNP筛选,并将基因型与兔各日龄体质量做关联性分析。【结果】测序结果经比对,在兔FSHR基因第10外显子发现突变g.163537 G>A和g.163619 A>G。其中g.163537 G>A为同义突变,处于低度多态(PIC<0.25),群体处于Hardy-Weinberg平衡;g.163619 A>G为错义突变,编码氨基酸由异亮氨酸变为缬氨酸,同样处于低度多态(PIC<0.25),群体处于Hardy-Weinberg平衡。对单位点基因型间体质量差异进行分析发现,g.163537 G>A位点对兔体质量影响不显著,但GG基因型个体的49、56及70日龄体质量高于GA和AA基因型个体,表明GG基因型增重速度较快;g.163619 A>G的AA基因型个体42、49日龄体质量显著高于AG基因型个体(P<0.05),尽管56、70日龄各基因型间差异不显著,AA基因型个体体质量仍然高于其他基因型。结果表明FSHR基因可以作为兔早期体质量性状选择的标记基因。[Objective]In recent years,extensive expression of FHSR has also been found in non-gonadal tissues,indicating that FSHR can mediate FSH signal to regulate many physiological activities in the body.FSH-FSHR signal has been shown to be related to fat deposition and body weight,but the relationship between FSHR gene and the body weight of rabbits has not been reported.[Methods]In the study,the PCR products of exon 10 of FSHR gene of Tianfu black rabbits was screened by DNA direct sequencing,and the correlation between genotype and the body weight of each day of rabbits was analyzed.[Results]Sequencing results were compared and two mutations(i.e.,g.163537 G>A and g.163619 A>G)in the exon 10 of FSHR gene were found.Among them,g.163537 G>A was a synonymous mutation with a low-level polymorphism information content(PIC<0.25)and the population was in Hardy-Weinberg equilibrium;g.163619 A>G was a nonsynonymous mutation resulting in the replacement of isoleucine by valine and exhibited a low-level polymorphism information content(PIC<0.25),and the population was also in Hardy-Weinberg equilibrium.The analysis of the difference between the genotypes of single mutation and body weight showed that g.163537 g>a locus had no significant effect on the body weight of rabbits,but the body weight of GG genotypes at 49,56 and 70 days was higher than that of GA and AA genotypes,indicating that the body weight of GG genotypes gained faster;AA genotypes of g.163619 A>G had significant higher body weight than that of AG genotype at 42 and 49 days(P<0.05),although there was no significant difference in the body weight of genotypes at 56 and 70 days,the body weight of AA genotypes was still higher than that of other genotypes.In conclusion,FSHR gene could be a candidate marker gene for early weight trait selection in rabbits.
