摘 要:目前阿尔茨海默病(AD)动物模型主要有氧化应激损伤、胆碱能损伤、铝中毒、β-淀粉样蛋白毒性和转基因等,其在造模周期、成功率等方面各具特点。本文通过整理近年建立AD动物模型的相关文献,以AD病因及病理机制为核心,总结并分析AD动物建模方法及评价指标,以期对AD模型进行合理的方法学评价。笔者认为,在现有AD动物模型的研究基础上,应结合新型的转基因技术和生物技术,构建与临床相符的AD模型症状,并参考AD临床诊断标准对动物模型的相关指标进行评价,将有利于全面评价AD模型。Alzheimer disease(AD)animal models mainly involve oxidative stress damage,cholinergic damage,aluminum poisoning,β-amyloid toxicity,and transgenic animals.These models have different characteristics that are mainly reflected in the modeling cycle and molding rate.By sorting out the related literature on AD modeling in recent years and focusing on the etiology and pathogenesis of AD,this paper summarizes and analyzes the methods of constructing AD models and evaluation indices,in order to make a proper methodological evaluation of AD models.It is believed that based on the research of existing AD models,new transgenic technology and biotechnology should be used to produce symptoms of the AD model that are consistent with the clinical practice.At the same time,the related indicators of animal models should be evaluated with reference to the clinical diagnostic standards of AD,which is conducive to comprehensive evaluation of AD models.
