摘 要:目的制备携带紫杉醇(paclitaxel,PTX)且能够在体外特异结合人卵巢癌细胞的纳米级脂质微泡,观察载药微泡特异度作用于OVCAR-3细胞及致其凋亡的能力。方法制备包裹PTX非特异度纳米微粒体(PTX-LNP)及携PTX特异度纳米微粒体(PTX-LNP-LHRH)。观察PTX-LNP-LHRH与PTX-LNP和二抗的结合情况;检测造影剂的靶连接率及不同药物浓度靶向载药组、非靶向载药组对卵巢癌细胞凋亡的影响。结果FITC标记的二抗与LHRH靶向微泡结合率高于非靶向组,约(96.53±0.82)%;体外寻靶实验示PTX-LNP-LHRH与OVCAR-3细胞紧密结合;流式检测LHRH靶向脂质载药微泡致OVCAR-3细胞的凋亡率显著高于PTX-LNP组,且其作用随药物浓度的增加而增强。结论PTX-LNP-LHRH可在体外与OVCAR-3细胞特异度结合,同时有效致靶细胞凋亡,且作用随着载药浓度的增加而增强。Objective To prepare Paclitaxel-loaded lipid microbubbles targeting ovcar-3 cells and observe their targeting effect on human ovarian cancer cells and ovcar-3 cell apoptosis.Methods Preparation of PTX-coated non-specific nanoparticles(PTX-lnP)and PTX-carrying specific nanoparticles(PTX-LnP-LHRH).To observe the combination of PTX-LNP-LHRH with PTX-LNP and secondary antibody.To detect the target connection rate of contrast agent and the effects of different drug concentrations on the apoptosis of ovarian cancer cells in targeted and non-targeted drug carriers.Results The binding rate of fitc-labeled secondary antibody to target group was as high as(96.53±0.82)%,In vitro targeting experiments showed that the targeted drug delivery microbubble contrast agent was closely bound to ovcar-3 ovarian cancer cells,The apoptosis rate of ovcar-3 cells in the targeted drug delivery group was significantly higher than that in the non-targeted group,and its effect was enhanced with the increase of drug concentration.Conclusions Paclitaxel loaded LHRH targeted lipid micro bubble contrast agent can efficiently target and bind ovcar-3 cells of human ovarian cancer in vitro and effectively induce target cell apoptosis.And its effect is enhanced with the increase of drug concentration.
