摘  要:目的探讨龙脷叶水提取物对哮喘模型大鼠肺组织病理学及血清中IL‐5、TXB2和6‐Keto‐PGF1α的影响。方法先采用腹腔注射卵清白蛋白(OVA)与氢氧化铝致敏、后雾化吸入OVA激发复制大鼠哮喘模型,通过HE染色观察大鼠肺组织病理改变,ELISA法检测血清IL‐5、TXB2和6‐Keto‐PGF1α浓度水平。结果与正常组比较,模型组支气管、血管周围和肺泡组织内炎性细胞可见明显增多,在血管周围浸润成环形成血管套袖,呈现明显哮喘病理改变;治疗组各部位炎性细胞明显减少,血管周围“套袖”样炎性浸润明显减少或消失,炎性病理改变明显减轻。与模型组比较,治疗组大鼠血清中IL‐5、TXB2浓度水平明显降低(P<0.01),6‐Keto‐PGF1α浓度明显升高(P<0.01),TXB2/6‐Keto‐PGF1α比值降低(P<0.05)。结论龙脷叶水提取物可以减轻哮喘模型大鼠肺组织与支气管炎性病理改变,推断可能是通过降低炎性因子IL‐5、TXB2/6‐Keto‐PGF1α浓度水平从而减轻炎症。Objective To explore the effect of water extract from Sauropus spatulifolius on lung histopathology and inflammatory factors IL‐5,TXB2 and 6‐Keto‐PGF1αin asthmatic rats.Methods The rat asthmatic model was induced by intraperitoneal injection of ovalbumin(OVA)and aluminum hydroxide,and then aerosol inhalation of OVA.The pathological changes of lung tissue were observed by HE staining.The serum levels of IL‐5,TXB2 and 6‐Keto‐PGF1 were detected by ELISA.Results Compared with the normal group,the inflammatory cells in the bronchi,perivascular and alveolar tissues of the model group significantly increased,and infiltrated into rings around the blood vessels to form vascular sleeves,showing obvious pathological changes of asthma.In the treatment group,the inflammatory cells in all parts were obviously reduced,the sleeves‐like inflammatory infiltration around blood vessels was obviously reduced or disappeared,and the inflammatory pathological changes were obviously alleviated.Compared with the model group,the levels of IL‐5,TXB2 and the ratio of TXB2/6‐Keto‐PGF1αwere significantly reduced in the treatment group,but the levels of 6‐Keto‐PGF1αwere markedly increased(P<0.05).Conclusion The water extract of Sauropus spatulifolius can alleviate the inflammatory pathological changes of lung tissue in asthmatic rats,which may be due to the modulation of inflammatory factors IL‐5,TXB2 and 6‐Keto‐PGF1α.
