摘 要:目的探究龙胆苦苷(gentiopicroside,GPS)对非小细胞肺癌(non‐small cell lung cancer,NSCLC)A549细胞的炎症因子水平、运动能力的作用。方法以非小细胞肺癌A549细胞系为研究对象,CCK8检测细胞活力,筛选合适的GPS浓度;选择GPS 0、5、10、20μmol/L分别对A549进行培养,收集细胞上清液,ELISA检测白细胞介素(interleukin,IL)‐6、诱导型一氧化氮合酶(inducible nitric oxide synthase,iNOS)和IL‐1β的水平;Transwell检测细胞侵袭,WB检测NF‐κBP65的磷酸化以及上皮细胞钙粘蛋白(E‐cadherin,E‐cad)、神经钙黏素(N‐cadherin,N‐cad)和波形蛋白(Vimentin)的蛋白表达;免疫荧光检测A549细胞中Vimentin的表达。结果GPS对A549细胞的活力有抑制作用;与GPS 0μmol/L相比,GPS(10和20μmol/L)可明显降低A549细胞上清液中IL‐6、iNOS和IL‐1β的含量及NF‐κBP65的磷酸化水平(P<0.05);明显减少A549细胞的侵袭数目(P<0.05);显著上调E‐cad蛋白表达(P<0.05),显著下调N‐cad和Vimentin蛋白表达(P<0.05)。结论GPS抑制非小细胞肺癌A549细胞的炎症因子产生、细胞侵袭和上皮间质转换,起到抑制肺癌的作用。Objective To explore the effect of gentiopicroside(GPS)on the levels of inflammatory factors and exercise capacity of non‐small cell lung cancer A549 cells.Methods CCK8 assay was used to detect cell viability and screen the appropriate GPS concentration.GPS at 0,5,10 and 20μmol/L were used to treat A549 cells,and the cell supernatant was collected.The levels of interleukin(IL)‐6,inducible nitric oxide synthase(iNOS)and IL‐1βwere detected by ELISA.Transwell was used to detect cell invasion.Western blot was employed to detect the phosphorylation of NF‐κB P65 and the expression ofE‐cadherin(E‐cad),N‐cadherin(N‐cad)and Vimentin.Immunofluorescence was commended to detect the expression of Vimentin in A549 cells.Results GPS had an inhibitory effect on the viability of A549 cells.Compared with GPS at 0μmol/L,GPS at 10 and 20μmol/L significantly reduced the contents of IL‐6,iNOS and IL‐1βin the supernatant of A549 cells and the phosphorylation level of NF‐κB P65(P<0.05).The invasion number of A549 cells was significantly reduced(P<0.05).The expression ofE‐cad protein was obviously increased(P<0.05),but the expression ofN‐cad and Vimentin protein was reduced(P<0.05).Conclusion GPS inhibits the production of inflammatory factors,cell invasion and epithelial mesenchymal transition of non‐small cell lung cancer A549 cells,which plays a role in inhibiting lung cancer.
