摘  要:目的:探讨尿石汤配方颗粒改善肾草酸钙结石(CaXO)大鼠肾功能效果及其机制。方法:建立草酸钙肾结石大鼠模型,检测大鼠造模前后体质量变化,肾脏质量、脏器系数变化,排尿量、尿液pH、尿液Ca^(2+)、尿液结晶数量及类型,观察肾结石模型在大鼠机体上的宏观变化。采用He染色和Von-Kossa’S染色,观察尿石汤配方颗粒对肾结石模型大鼠肾组织的病理变化。采用试剂盒方法,检测大鼠肾组织中SOD、MDA、GHS-PX、CAT指标变化,探讨尿石汤配方颗粒对改善肾草酸钙结石大鼠肾功能可能的机制。结果:模型组体质量增长缓慢,肾脏质量增加,肾脏系数升高,排尿量减少,pH呈碱性,Ca^(2+)浓度升高,BLD结果显示+3,PRO结果显示+2,尿液中结晶为磷酸铵镁结晶,结晶数量为+3,肾脏病理观察可见明显肾小管扩张和草酸钙晶体沉积。给药组体质量增长较模型组快,肾脏质量和肾脏比重较模型组不同程度下降,排尿量增多,pH呈碱性,Ca^(2+)浓度呈下降趋势,BLD结果显示+3,PRO结果显示阴性,尿液结晶数不同程度下降,肾脏病理观察可见肾小管扩张程度下降,草酸钙晶体减少。模型组大鼠肾组织中SOD、GSH-PX、CAT等指标水平下降,而MDA指标水平升高。各给药组均能提高因造模引起的SOD、GSH-PX、CAT水平降低,同时降低因造模引起的MDA水平升高。结论:尿石汤配方颗粒对改善肾草酸钙结石大鼠肾功能具有良好的治疗效果。改善草酸钙结石肾功能作用可能的机制为:阻止MDA的产生,减少细胞损伤,激活SOD、GSH-PX、CAT的释放,以防止生成CaXO结晶。OBJECTIVE To explore the efficacy and mechanism of urine stone decoction granules for improving renal function of rats with calcium calcium oxalate calculi(CaXO).METHODS Calcium oxalate nephrolithiasis rat model was established for detecting the weight change of rats before and after modeling. The change of kidney weight, organ coefficient, urine output, urine pH, urine Ca^(2+) and urine crystal quantity/type were recorded. Various kits were employed for detecting the changes of SOD, MDA, GHS-PX and CAT in rat kidney tissues and elucidating the possible mechanism of Niaoshi Tang formula granules for improving renal function.RESULTS In model group, weight gain was slow, kidney weight gained, kidney coefficient increased, urine output decreased, pH became alkaline and Ca^(2+) concentration increased. BLD results showed +3, PRO results +2 and urine crystals were magnesium ammonium phosphate. The number of crystals was +3. Renal pathology revealed obvious renal tubular dilation and calcium oxalate crystal deposition. The weight gain of drug administration group was faster than that of model group. Kidney weight and specific gravity of kidney decreased, urine output increased, pH became alkaline and Ca^(2+) concentration showed a downward trend. BLD result showed +3, PRO result negative, urine crystals decreased and renal pathology indicated that the degree of renal tubular dilation decreased and calcium oxalate crystals decreased in model group, the levels of SOD, GSH-PX, CAT and other indicators in kidney tissue decreased while the levels of MDA increased. The levels of SOD, GSH-PX and CAT increased and the level of MDA decreased after modeling.CONCLUSION Formula granules of Niaoshi Tang are efficacious for improving renal function of rats with calcium calcium oxalate stones. Possible mechanisms are preventing the production of MDA, reducing cell damage and activating the releases of SOD, GSH-PX and CAT for preventing the formation of CaXO crystals.
