摘  要:为了提高薄皮甜瓜的果皮花斑和非果皮花斑性状育种材料的选择效率,笔者采用田间提供的薄皮甜瓜的果皮花斑材料和非果皮花斑材料进行了研究,于2号染色体上控制甜瓜果皮花斑的基因附近重新开发连锁的InDel-spot分子标记,利用InDel-spot标记对20份薄皮甜瓜材料进行基因检测。结果表明,果皮花斑条带大小约211 bp,而果皮非花斑条带大小约219 bp,且符合率达到95%;进一步利用InDel-spot标记对2个F2群体及5个F3群体进行果皮花斑的鉴定,其中Qb978(F2)、Qb931(F3)和Qb932(F3)的检测准确率分别96.6%、91.7%和83.3%。该InDel-spot分子标记在甜瓜果皮花斑的实际鉴定中具有重要的实践运用,可以针对性地提高育种材料选择的效率,缩短育种周期。The purpose is to improve the selecting efficiency of breeding materials with the spotted and the non-spotted trait in melons. The authors conducted the research with the spotted and non-spotted melons provided in field, and developed the linked InDel-spot molecular marker near the gene controlling the melon spotted trait in chromosome 2. Twenty melons were genotyped with the InDel-spot maker. The results showed that the size of spotted band was about 211 bp, while the non-spotted band was about 219 bp, with 95% of compliance rate. The InDel-spot marker was further used to identify the spotted and non-spotted trait of 2(F2)and 5(F3) populations. Among them, the detection accuracy of Qb978(F2), Qb931(F3) and Qb932(F3) was96.6%, 91.7% and 83.3%, respectively. The results indicate that InDel-spot marker has a great application value in the identification of the spotted and non-spotted trait, which could raise the selecting efficiency of breeding materials and shorten the breeding cycle.
